摘 要:目的观察周期性单轴牵张力对前软骨干细胞增殖能力的影响。方法免疫磁珠分选新生大鼠前软骨干细胞(precartilaginous stem cells,PSCs),取生长良好的第三代PSCs,通过四点弯曲细胞加压装置对细胞加载不同的机械牵张力(2 000、4 000μstrain),并在不同时间点(1、2、4和8 h)收集细胞,采用流式细胞仪检测应力刺激后PSCs的细胞周期及增殖指数的变化,揭示不同的牵张力在不同的时间点对PSCs增殖活性的影响。结果周期性单轴牵张力能影响大鼠PSCs的增殖活性。2 000μstrain机械牵张力组与对照组相比,增殖指数升高(P<0.05),促进细胞增殖,并且S期DNA合成率增多(P<0.05);而4 000μstrain机械牵张力组与对照组比较,增殖指数降低,抑制细胞增殖(P<0.05),同时S期细胞DNA合成率减少(P<0.05)。结论适宜的机械牵张力能促进PSCs增殖,过大的机械牵张力反而抑制细胞增殖。Objective To investigate the proliferation of the rat precartilaginous stem cells(PSCs) in response to different uniaxial cyclic tensile stretch.Methods The rat PSCs were isolated and purified using the magnetic activated cell sorting and cultured in vivo.The PSCs at third generation were stimulated by a four-point bending system at different tensile stretch(2 000,4 000 μstrain) and different time points(1,2,4 and 8 h).Flow cytometry was used to explore the changes of cell cycle and proliferation index(PI).Results Tensile stretch could influence the proliferation of PSCs.As compared with the control group,the 2 000-μstrain mechanical stretch could promote the proliferation of PSCs,with increased PI(P0.05) and the synthesis ratio of DNA(P0.05) in S phase.But 4 000-μstrain mechanical stretch could inhibit the proliferation of PSCs,with decreased PI(P0.05) and the synthesis ratio of DNA(P0.05) in S phase.Conclusion Proper tensile stretch could facilitate the proliferation of PSCs,while overloaded tensile stretch could inhibit it.
