摘 要:目的:研究梯度热打击对体外培养骨骼肌细胞的损伤和表达IL-6、TNF-α的影响。方法:利用CCK-8法对比各组不同时相细胞存活率和24h增殖率。采用ELISA法对比梯度热打击量对骨骼肌细胞释放IL-6和TNF-α的影响。结果:对比37℃对照组,热打击组各个时相细胞活力均下降(P<0.001),并呈时间及温度依赖关系。对比37℃对照组,39℃组7h时段及41℃和43℃组各时相增殖率显著下降(P<0.001),并呈时间及温度依赖关系。ELISA检查显示,热打击组IL-6和TNF-α的表达量较37℃对照组显著上升(P<0.001),41℃和43℃组较39℃组显著上升(P<0.01)。结论:热打击对骨骼肌细胞具有细胞毒效应,抑制骨骼肌细胞的增殖,并促进细胞因子IL-6和TNF-α的释放。Objective To investigate the effect of gradient heat stress on skeletal muscle cells injuries and release of IL-6 and TNF-α.Methods CCK-8 assay was used to investigate the difference of cell survival rate and 24 h proliferation rate among each group.ELISA was used to investigate the difference of IL-6 and TNF-α release in each group.Results In comparison with normal control group,the cell vitality of heat stress group were significantly decreased in each phase(P 0.001),and this decrease appeared time-and thermal gradient-dependent.The proliferation rate of 7 h phase of 39℃ group and each phase of 41℃ and 43℃ groups were significantly decreased in comparison with normal control group(P 0.001).This decrease was also time-and thermal gradient-dependent.The expression level of IL-6 and TNF-α of heat stress groups were significantly up-regulated in comparison with normal control group(P 0.001).More IL-6 and TNF-α were released in cells of 41℃ and 43℃ groups than in cells of 39℃ group(P 0.01).Conclusion Heat stress has cytotoxic effect on skeletal muscle cells,and can inhibit proliferation ability of these cells.Heat stress can promote the IL-6 and TNF-α release of these cells.
