摘 要:目的研究红霉素(ERY)对铜绿假单胞菌QS调控毒力因子分泌的干预作用。方法弹性蛋白-刚果红法测定弹性蛋白酶活性;偶氮酪蛋白法测定蛋白水解酶活性;氯仿萃取法测定绿脓菌素;苔黑酚法测定鼠李糖脂;测定早期及成熟生物被膜细菌(BF)对H2O2敏感性。结果 ERY组弹性蛋白酶活性(0.03)、蛋白水解酶活性(0.37)、绿脓菌素浓度(0.06)、鼠李糖脂浓度(48.8μg/ml)均明显低于空白对照组(分别为:0.47、0.48、0.18、153.9μg/ml)(P<0.01),弹性蛋白酶活性、蛋白水解酶活性及绿脓菌素浓度高于PA-JP3株组(分别为:0.04、0.16、0.01)(P<0.05),而鼠李糖脂浓度与PA-JP3株组(34.8μg/ml)相当,ERY组(早期1.81cm,成熟期3.8cm)及PA-JP3株组(早期2.6cm,成熟期2.87cm)生物被膜细菌对H2O2敏感性高于空白对照组(早期1.36cm,成熟期1.44cm)(P<0.01)。结论红霉素可以抑制铜绿假单胞菌毒力因子的产生。OBJECTIVE To evaluate the intervention effect of erythromycin on Pseudomonas aeruginosa PAO1 QS-controlled virulence factors.METHODS LasB elastolytic activity was determined by using Elastin-Congo Red.Protease activity was determined by using Azo-casein.Chloroform extraction method was used for pyoverdine assay.The orcinol assay was used to directly assess the amount of rhamnolipids.The H2O2 susceptibility of the young and mature BF bacterial was determined.RESULTS The virulence factors production in EM group(LasB:0.03,protease:0.37,pyoverdine: 0.06,rhamnolipids:48.8ug/ml) were significantly lower than that in the control group(0.47,0.48,0.18,153.9 μg/ml,respectively)(P0.01),the rhamnolipids were even reduced to the same as with the PA-JP3 group(34.8ug/ml),but LasB elastolytic activity,pyoverdine and the protease activity were higher than the PA-JP3 group(LasB:0.04,protease:0.16,pyoverdine:0.01)(P0.05).Both the young and the mature BF bacterial in EM group(1.81cm and 3.8cm) and the PA-JP3 group(2.6cm and 2.87cm) were more susceptible to H2O2 than the control group(1.36cm and 1.44cm)(P0.01).CONCLUSION Erythromycin can inhibit the production of PAO1 virulence factors.
