摘  要:目的比较苦味酸-天狼猩红染色法和MASSON染色法在评价小鼠心脏纤维化中的异同。方法 10周龄BALB/c雄性小鼠给予皮下注射异丙肾上腺素(5mg/kg/d)共14天,诱导小鼠心脏纤维化模型,取心脏标本制备石蜡切片,分别行苦味酸-天狼猩红染色和MASSON染色,观察胶原纤维的沉积。结果苦味酸-天狼猩红染色法和MASSON染色法都能清楚地显示出胶原纤维,且其定位是一致的。两种染色方法显示出的纤维化面积,在统计学上无显著性差异(P>0.05)。但苦味酸-天狼猩红染色法处理的切片能够在偏振光显微镜下清楚地区分Ⅰ型和Ⅲ型胶原纤维,而MASSON染色法却不能。结论苦味酸-天狼猩红染色法较MASSON染色法相比能够更好地评价心脏纤维化。Objective To compare picric Sirius red staining and MASSON staining method in evaluation of mouse cardiac fibrosis.Methods 10 week old BALB/C male mice were given subcutaneous injection of isoproterenol(5mg/kg/d) for 14 days,which induced cardiac fibrosis model successfully.In order to observe the collagen deposition,we prepared paraffin sections by using the mouse heart specimen,and carried out picric Sirius red staining and MASSON staining respectively.Results Both picric Sirius red staining and MASSON staining method could clearly show the collagen fibers,and similar localization.There was no significant difference on the fibrosis area(P0.05).However,under polarized light,we could clearly distinguished the type I and type III collagen fiber by using picric Sirius red staining but not MASSON staining.Conclusion Picric Sirius red staining is better than MASSON staining to be used for the evaluation of cardiac fibrosis.
