摘 要:目的研究黄芩苷对人肺泡上皮细胞的抗氧化作用。方法用过氧化氢制作肺泡Ⅱ型上皮细胞A549氧化损伤模型,再加入不同浓度的黄芩苷进行处理,MTT法测定细胞活力、测定细胞培养上清液中谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)的活性。结果氧化损伤组的细胞活力显著降低、MDA水平显著上升、S0D、GSH-PX、NOS活性下降(P<0.05)。黄芩苷处理各组与损伤组比较,细胞活性显著升高、MDA水平显著下降、S0D、GSH-PX、NOS活性和蛋白含量显著上升(P<0.05)。结论黄芩苷对过氧化氢引起的A549细胞氧化损伤具有保护作用。Objective To investigate the baicalin's protective effect on oxidation damage in A549.Methods A549 cells were treated with H2O2 as oxidative stress damage cell model and different concentrations of baicalin were added.Cell viability was evaluated by MTT assay and the level of MDA and SOD、NOS、GSH-PX in cell culture supernatant was measured with antioxidation testing cassete.Results The MTT assay results showed that H2O2 decreased the viability of A549 cells(P0.05),the protein level decreased and oxidative damage index rose on the group of H2O2.Baicalin inhibited the decrease of A549 cell viability and protein induced by H2O2(P0.05).The decrease of SOD、NOS、GSH-PX was improved(P0.05).The viability of SOD,NOS,GSH-PX and protein content increased significantly.Conclusions Bacalin could inhibit the injury caused by H2O2 and reduce the oxidative damage effect induced by H2O2(P0.05).
