摘  要:目的:观察黄芪甲苷干预对急性肺损伤大鼠肺组织水通道蛋白(AQP1)表达的影响,并探讨其可能的机制。方法:SPF级雄性SD大鼠63只,随机分为模型组(acute lung injury,ALI组)、正常对照组(normal saline,NS组)、黄芪甲苷干预组(astragaloside,AS组)。ALI模型复制方法为舌下静脉注射LPS 4 mg.kg-1;注射前AS组给药0.6 mg.kg-1 AS连续灌胃7d,ALI组给予NS7 d,NS组不做任何灌胃处理,7 d后舌下给药NS。观察注射后2,6,12 h肺组织湿干重(W/D)、肺组织病理学改变、支气管肺泡灌洗液(BALF)的细胞总数及中性粒细胞百分比、免疫组化与Western blot法肺组织AQP1蛋白表达的变化。结果:ALI组各个时点W/D值、BALF细胞总数及中性粒细胞百分比升高较NS组上升,AQP1、蛋白表达较ALI组下降,以6,12 h最明显,ALI组与NS组各时点间的差异均具有统计学意义(P<0.05)。AS组各个时点W/D值、BALF细胞总数及中性粒细胞百分比升高较ALI组降低,AQP1、蛋白表达较ALI组上调,以6,12 h最明显,具有统计学意义(P<0.05)。结论:AS能减轻LPS致大鼠肺炎症损伤反应,其机制与上调肺AQP1的表达有关。OBJECTIVE To explore the protective effect and mechanism of action of astragaloside (AS) on acute lung injury (ALl) rat induced by LPS. METHODIS 63 SPF male SD rats were randomly divided into 3 groups as following: control group, model group, and astragaloside group (AS group), with 21 rats in each group respectively. The model group was treated with normal saline 0. 2 mL.d 1 orally for 7 days, in AS group, 4. 5% astragaloside (0. 6 mg.kg-1.d-1 , the concentration of 4. 5 mg-mL-1 ) was administered by i. g. for 7 days, and each rat in control group was given the normal saline 0. 2 mL-d-1 orally. The model was reproduced by sublingual intravenous injection of LPS 4 mg. kg-1 excepet the control group which was treated with NS by sublingual intravenous injection. Dynamic observation in each group was done on the change of wetdry weight (W/D), histopathological changes in lung tissue, bronchoalveolar lavage fluid (BALF) total cells and percentage of neutrophils, immunohistochemical and Western blotting method lung tissue AQP1 protein 2,6 h, and 12 h after injection respectively. RESULTS Compared with those in the NS group, ALI group W/D values, BALF total cells and neutrophil per-centage 2 h after intravenous injection of LPS began to increase, and reached the peak at the 6th hour and the 12th hour; lung tissue AQP1 protein expression decreased 2 h after intravenous injection of LPS; and at the 6th hour and the 12th hour reduced significantly. The differences between ALl group and NS group at all time points are statistically significant (P〈0. 05). AS group at various time points, the W / D values, BALF total cells and neutrophil percentage increase were lower than ALI group, AQP1 expression in ALI group were on the increase , with the 6th hour and the 12th hour the most obvious, and they were of statistical significance (P〈0. 05).
