莫达非尼R-和S-对映体的LC-MS/MS定量检测以及在Caco-2单层细胞模型上的转运特性

摘　　要：目的建立准确、灵敏、可靠的LC-MS/MS法,定量测定莫达非尼对映体,考察其在Caco-2细胞单层模型上的转运。方法选用资生堂C8(50 mm×2.1 mm,3μm)色谱柱及API4000液相色谱-质谱联用仪,用于定量分析的离子反应分别为m/z 274.4→165.1(莫达非尼)和m/z 391.2→265.1(CS55)。结果在本实验条件下,建立范围为1～1000 ng/ml的定量方法,批内、批间精密度<15%(n=5)。结论建立一种快速、准确、灵敏度高、专属性好,可用于莫达非尼对映体在Caco-2单层细胞模型转运研究的方法。Objective To establish a highly sensitive,rapid and selective liquid chromatography mass spectrometry（LC-MS/MS） method for the quantitation of modafinil enantiomers and the analysis of enantiomers＇ transport characteristics in a Caco-2 monolayer model.Methods An API4000 mass spectrometer was applied as the detector and using the precursor ion to product ion（m/z 274.4 to 165.1 for modafinil and 391.2 to 265.1 for CS55） was performed to quantify the modafinil.Results The range of the calibration curve was 1-1000 ng/ml.The intra-and inter-run precision RSD was less than 15%.Conclusion This LC-MS/MS method is sensitive,simple,rapid and suitable for determination of modafinil enantiomers.A Caco-2 monolayer model is successfully established to study the transport characteristics of modafinil enantiomers.
