摘  要:目的研究大气PM2.5对人支气管上皮细胞(16-HBE)DNA的损伤效应。方法将人支气管上皮细胞(16-HBE)分别暴露于8、16、32、64、128μg/ml的广州大气PM2.5 24、48、72 h,采用MTT法测定细胞存活率,彗星试验检测细胞DNA损伤水平。结果染毒24、48、72 h后,64、128μg/ml组细胞存活率均明显低于对照组(P<0.05),各染毒组细胞Olive尾矩均明显高于对照组(P<0.05);72 h各剂量组细胞存活率均低于24 h相应处理组(P<0.05),48、72 h各剂量组细胞Olive尾矩与24 h相比均明显增加(P<0.05);经多重线性回归分析,Olive尾矩(y)与染毒剂量(d)和染毒时间(t)的回归方程为y=-6.481+0.162 d+0.210t(R^2=0.842,P<0.05)。结论大气PM2.5可导致人支气管上皮细胞(16-HBE)DNA损伤,且损伤程度与染毒剂量、时间有一定关联。Objective To study the effect of airborne particulate matter PM2.5 on DNA damage in human bronchial epithelial cells(16-HBE). Methods The human bronchial epithelial cells(16-HBE) were exposed to 8, 16, 32, 64, 128 μg/ml airborne particulate matter PM2.5 collected from Guangzhou. After 24,48 and 72 h of incubation, the cell survival was measured by MTT assay. DNA damage was detected with Comet assay. Results After 24, 48 and 72 h of treatment, compared with the negative control, the cell survival rates of 64, 128μg/ml groups decreased significantly (P〈0.05), and the DNA Olive tail moment of all groups increased significantly (P〈0.05). Compared with the 24 h group, the cell survival rates of all treated groups in 72 h decreased significantly (P〈0.05), and the DNA Olive tail moment of all groups in 48 and 72 h increased significantly (P〈0.05). The regression equation between the DNA Olive tail and dose-time is y=-6.481+0.162 dose+0.210 time (R2=0.842,P〈0.05). Conclusion Airborne particulate matter PM2.5 can induce human bronchial epithelial cells (16-HBE) DNA damage in a dose-time dependent manner.
