摘 要:目的:检测和控制牛肾细胞基质中可能存在的外源病毒,提高细胞基质的安全性,保证轮状病毒疫苗的质量。方法:用ELISA法检测牛肾供体新生小牛血清中的病毒抗体。用细胞培养法检测供体血清和牛肾细胞中的外源病毒,免疫荧光抗体法检测供体血清和牛肾细胞中的特殊牛源病毒。结果:81份禁食禁水的新生小牛血样7种病原抗体检测均为阴性;血吸附病毒检测均为阴性,直接免疫荧光检出1例BVDV阳性。65份牛肾细胞特殊牛源病毒检测均为阴性。结论:通过对牛肾供体小牛进行病原筛选可有效降低牛肾细胞携带外源病毒的风险,也是提高疫苗安全性行之有效的策略。Objective: To detect and control potential adventitious viruses in bovine kidney cell substrates and to improve the safety of the cell substrate for the quality assurance of the rotavirus vaccine. Methods: ELISA methods were used to detect antibodies against the viruses in serum derived from kidney donors, new born calves. Cell culture techniques followed by virus-specific fluorescent detection assay were applied to examine the presence of bovine pathogens. Result : None of 81 serum samples from new born calves without feeding was positive with seven bovine virus antibodies, the hemadsorption test have same result,one was BVDV positive by direct fluorescent antibody assay. All of 65 cell samples were free of bovine viruses that examined. Conclusion : The risk of contamination with adventitious viruses in bovine kidney cell substrate can be effectively minimized by examining the pathogens of donor calves. It can be a efficient mitigation strategy for vaccine safety.
