摘 要:目的:评估川芎嗪对长时间丙泊酚所致遗忘小鼠的记忆改善效果。方法:昆明种小鼠90只,体重17~25g,雌雄各半。按分层随机区组设计,将小鼠分成6组,分别为:生理盐水组(NS组)、10mg.kg-1丙泊酚组(P10组)、7mg.kg-1川芎嗪组(L7组)、20mg.kg-1川芎嗪组(L20组)、7mg.kg-1川芎嗪+10mg.kg-1丙泊酚组(L7+P10组)、20mg.kg-1川芎嗪+10mg.kg-1丙泊酚组(L20+P10组),每组15只。各组小鼠单独或联合腹腔内注射不同剂量的丙泊酚、川芎嗪、或生理盐水,日1次,连续9天。于实验第10天,即连续给药后第1天,行跳台训练。并于训练后24h、48h及72h,分别测试小鼠的学习记忆能力。结果:(1)小鼠训练后24h、48h测验,与NS组相比,P10组小鼠潜伏期缩短(P<0.05)、错误次数增多(P<0.05);训练后72h测验,P10组小鼠的潜伏期、错误次数与NS组相似(P>0.05);(2)小鼠训练后24h、48h、72h测验,与NS组相比,L7组、L20组小鼠潜伏期、错误次数均相似;(3)小鼠训练后24h、48h测验,与P10组相比,L7+P10组、L20+P10组小鼠均表现为潜伏期延长(P<0.05)、错误次数减少(P<0.05);训练后72h测验,L7+P10组、L20+P10组与P10组的潜伏期、错误次数均相似(P>0.05)。结论:日1次,连续9d,腹腔注射10mg.kg-1丙泊酚可致小鼠遗忘;7mg.kg-1或20mg.kg-1川芎嗪与10mg.kg-1丙泊酚合用,均能有效改善后者所致的小鼠记忆障碍。Objective: To investigate the effect of long-term administration of propofol on memory function and the protective effect of ligustrazine on memory function in propofol-induced amnesia in mice.Methods: We used step-down test in this experiment.90 Kunming mice,17g^25g,were divided randomly into 6 groups,15mice in each group.Mice in group NS,group P10,group L7,group L20,group L7+P10 and group L20+P10 were administrated normal saline of 10mL kg-1,propofol of 10mg·kg-1,ligustrazine of 7mg·kg-1,ligustrazine of 20mg·kg-1,7mg·kg-1 ligustrazine and 10mg·kg-1 propofol,20mg·kg-1 ligustrazine and 10mg kg-1 propfol by intraperitoneal injection once per day for 9 days,respectively.Step-down test was operated after 9-day-term administration finished.We tested the memory of mice at 24 hours,48 hours and 72 hours later after the training,respectively.The markers of memory 7s based on the latencies and the numbers of errors.Results:(1) Group P10 compared with group NS,the latencies were shorter and the numbers of errors were increased at 24h and 48h post-training(P0.05).At 72h post-training,the latencies and the numbers of errors were not statistically significant in group NS and group P10.(2) Compared with group NS,the latencies and the numbers of errors were not statistically significant in group L7 and group L20 at 24h,48h and 72h post-training,respectively.(3) At 24-h and 48-h post-training,compared with group P10,the latencies were longer and the numbers of errors were decreased in group L7+P10 and group L20+P10,respectively(P0.05).At 72-h post-training,the latencies and the numbers of errors were not statistically significant in group NS,group L7+P10 and group L7+P10.Conclusions: These results indicated that 9-day-term administration of 10mg·kg-1propofol can induces amnesia,and 7mg·kg-1 or 20mg·kg-1 ligustrazine can prevent propofol-induced amnesia in mice.
