慢盆消炎方对慢性子宫内膜炎模型大鼠NF-κB、IκB-α及TNF-α、MCP-1表达的影响

摘　　要：目的:观察慢盆消炎方对慢性子宫内膜炎模型大鼠NF-κB、磷酸化IκB-α、TNF-α、MCP-1的表达及其TNF-α、MCP-1含量变化的影响。方法:采用混合菌株接种法制作慢性子宫内膜炎大鼠模型,将造模成功的大鼠随机分为:模型组、妇乐组、中药组(慢盆消炎方),另设同龄正常大鼠为正常组,连续灌胃给药3周。给药结束取各组大鼠子宫内膜组织进行病理形态学观察;Western blot分析NF-κB、磷酸化IκB-α蛋白在子宫内膜组织表达水平;RT-PCR法检测各组子宫内膜组TNF-α、单核细胞趋化蛋白-1(MCP-1)mRNA表达;ELISA法测定各组大鼠血清TNF-α、MCP-1含量。结果:1、与正常组相比,模型组子宫内膜组织NF-κB蛋白、磷酸化IκB-α蛋白、TNF-αmRNA、MCP-1 mRNA表达水平显著上调(P<0.01或P<0.05),血清中TNF-α、MCP-1含量亦明显上升(P<0.01);病理学显示模型组子宫内膜呈炎症改变,各层组织均有大量的炎性细胞浸润,并且黏膜层有出血。2、与模型组相比,妇乐组子宫内膜组织NF-κB蛋白、磷酸化IκB-α蛋白、TNF-αmRNA、MCP-1 mRNA表达水平有所下降(P<0.05),但下降程度不如中药组;病理学显示上皮细胞结构已有较明显的修复,排列较整齐,但子宫内膜仍可见少量淋巴细胞浸润,伴腺体轻度扩张。3、与模型组相比,中药组子宫内膜组织NF-κB蛋白、磷酸化IκB-α蛋白、TNF-αmRNA、MCP-1 mRNA表达水平均显著下调(P<0.01),血清中TNF-α、MCP-1含量亦明显下降(P<0.01);病理学显示中药组子宫内膜各层结构基本恢复正常,仅见极少量淋巴细胞浸润。结论:慢盆消炎方具有治疗子宫内膜炎的功效,其机制可能与抑制NF-κB、磷酸化IκB-α、TNF-α;MCP-1表达及降低TNF-α、MCP-1含量相关。Objective：To observe the expressions of NF-κB、phospho-IκB-α、TNF-α、MCP-1 and the serum concentrations of TNF-α and MCP-1 in chronic endometrial inflammatory rats treated with Manpan Xiaoyan Formula.Methods：Rats were infected with mixed inocula of Escherichia coli,Staphylococcus and Streptococcus pyogenes to design a model of chronic endometritis rats.Endometrial tissues in rats were examined for histopathology.The successful model rats were randomly divided into： Model group,Fule group,traditional Chinese medicine group（Manpan Xiaoyan Formula）,the other normal rats were identified as normal group.These rats were continuously irrigated stomach with medicine for three weeks.The expressions of NF-κB and phospho-IκB-α in endometrial tissues were detected by western blot.The expressions of TNF-α and MCP-1 mRNA in endometrial tissues were examined by RT-PCR.The serum concentrations of TNF-α and MCP-1 in rats were detected by ELISA.Results：1.Compared with control group,the expressions of NF-κB、phospho-IκB-α、TNF-αmRNA and MCP-1 mRNA in endometrial tissues of model group were obviously increased（P0.01、P0.05）.The serum concentrations of TNF-α and MCP-1 in model group were also significantly higher（P0.01）.Histopathology showed endometrial tissues were changed by inflammation,and layers of endometrial tissues were infiltrated by inflammatory cells and mucosa bleeding.2.Compared with model group,the expressions of NF-κB、phospho-IκB-α、TNF-αmRNA and MCP-1 mRNA in endometrial tissues of model group were decreased（P0.05）.However,the extent of the decline was less than traditional Chinese medicine group.Histopathology showed the structure had been more pronouncedly repaired epithelial cell,arranged in neat rows,but still showed a small amount of endometrial lymphocytes infiltration,with mild expansion of the glands.3.Compared with model group,the expressions of NF-κB、phospho-IκB-α、TNF-αmRNA and MCP-1 mRNA in endometrial tissues of Chinese medicine group were obvious
