鸡大肝和大脾病抗原在免疫器官的定位和分布

摘　　要：用免疫金 银法 (IGSS)和免疫胶体金细胞化学方法 ,对人工感染的大肝和大脾 (BLS)病鸡免疫器官作BLS抗原(BLSAg)定位、分布及其动态研究。结果感染后第 1,7,19周 ,在脾脏、盲肠扁桃体、胸腺、法氏囊、红骨髓和哈德尔氏腺组织中均检查到BLSAg ,主要分布在淋巴细胞、网状细胞、网状上皮细胞、巨噬细胞、单核细胞、浆细胞的胞浆、内质网、线粒体和一些空泡内。BLSAg阳性淋巴细胞早期主要分布在胸腺和外周免疫器官的T 淋巴细胞区 ,第 7周在胸腺、法氏囊和外周免疫器官的T、B淋巴细胞区。结果初步表明BLS病毒定位于胞浆。机体在抗BLS病毒方面 ,早期以体液免疫为主 ,而中期的体液免疫和细胞免疫均可能不同程度的降低。Location and the distribution of BLS antigen(BLSAg)were dynamically observed from the immune organs of broiler breeders experimentally inoculated with BLS disease agent by the methods of immuno gold silver staining(IGSS)and immuno colloidal gold cytochemistry.The BLSAg appeared in the spleen,the tonsillia cecalia and thymus,the bursa Fabricius,the red marrow and Harderian's gland at the lst week,7th week and 19th week post inoculation(P.I.).The BLSAg mainly located in the cytoplasm,endoplasmic reticulum,the mitochondria and some vacuole of the lymphocytes,reticulocytes and the epithelia reticular cell,the macrophages,the monocytes and plasma cells.The BLSAg positive lymphocytes mainly distributed in the thymus and the areas of the T lymphocytes in the periperal immune organs at the early stage,and they appeared extensively in the thymus,bursa Fabricius and the area of T and B lymnhocytes in the periperal immune organ at the 7th week P.I..The results indicated that the BLS virus located in the cytoplasm.This also showed that the humoral immunoresponse(HI)played an important role at the early stage,and that both of HI and the cell mediated immunoresponse may be decreased in varying degree at the 7th week.
