扶正软坚散结法对甲减大鼠心肌α-MHC和β-MHCmRNA表达的影响

摘　　要：目的:研究扶正软坚散结法对甲状腺功能减退大鼠心肌肌球蛋白重链α和β(简称α-MHC和β-MHC)mRNA表达的影响。方法:将甲减Wistar大鼠随机分为模型对照组、L-T4组、芪贝复元饮组。正常对照组:普通饮食,双蒸水;模型对照组:低碘饮食,双蒸水。双抗体一步夹心法酶联免疫吸附法(ELISA法)测大鼠血清促甲状腺激素(TSH),放射免疫分析法(RIA法)测大鼠血清总T3(TT3)、总T4(TT4)。砷铈分光光度法测定尿碘。光镜下观察心肌细胞形态。实时定量RT-PCR法测心肌组织α-MHC和β-MHC mRNA的表达。结果:56天(8周)时,与模型组比较,予处理因素的L-T4组和芪贝复元饮组大鼠血清TT3值均升高(P<0.01),但两组之间比较无差异;血清TT4值均升高(P<0.001),二组之间比较无差异;TSH值明显降低(P<0.01),两组之间比较有差异(P<0.05),芪贝复元饮组优于L-T4组。L-T4组、芪贝复元饮组的α-MHCmRNA的表达分别为正常组的1.77倍、2.03倍,与模型组比均有上升,芪贝复元饮组较L-T4组上升明显;L-T4组、芪贝复元饮组β-MHCmRNA的表达分别为正常组的2.61倍、1.22倍,与模型组比均有下调,均未降低至正常水平,芪贝复元饮组下调明显。结论:扶正软坚散结法在甲减心肌损伤的修复中起着重要的作用,与提高心肌α-MHCmRNA的表达,降低β-MHCmRNA的表达有重要的关系。Objective：To study the effect of Fuzheng Ruanjian Sanjie method on the expressions of mRNA of hypothyroid rats myocardial myosin heavy chain alpha and beta.Methods：Hypothyroid Wistar rats were randomly divided into model group,L-T4 group,Qibei Fuyuan Decoction group.Normal group：ordinary diet,double steamed water;model group：low iodine diet,double steamed water.Serum thyroid-stimulating hormone（TSH）was measured with double antibody step clip method and ELISA method（ELISA）,serum total T3（TT3）and total T4（TT4）were measured with radioimmunoassay（RIA）.Ger-Arsenite color metric method was used to measure median urinary iodi（MUI）.Myocardial cells forms were observed under Light microscopy.The expressions of mRNA of myocardial myosin heavy chain alpha and beta were measured with Real-time RT-PCR.Results：56 days later,compared with model group,serum TT3 value of L-T4 group and Qibei Fuyuan Decoction group increased significantly（P0.01）,no discrepancies existed between the two groups;serum TT4 value was increased（P0.001）,no discrepancies existed between the two groups;TSH value reduced significantly（P0.01）,there were discrepancies between the two groups（P0.05）,Qibei Fuyuan Decoction group was better than L-T4 group.The expressions of a-MHC mRNA of L-T4 group and Qibei Fuyuan Decoction group were respectively 1.77 times and 2.03 times higher than that of normal group,compared with model group,they were increased,Qibei Fuyuan Decoction group increased obviously;the expressions of β-MHC mRNA of L-T4 group and Qibei Fuyuan Decoction group were respectively 2.61 times and 1.22 times higher than that of normal group,compared with model group,they were reduced,but all did not drop to normal levels,Qibei Fuyuan Decoction group reduced obviously.Conclusion：Fuzheng Ruanjian Sanjie method plays an important role on hypothyroid rats myocardial injury repaired.It has important relationship with improving the alpha MHCmRNA expression and reducing the beta MHCmRNA expression.
