摘 要:研究4种基本培养基和3种剑麻外植体对愈伤组织诱导及分化的影响。结果表明,SH基本培养基对剑麻愈伤组织的诱导和分化效果最好,出愈率为100%,愈伤量大,分化率为21.1%;用于剑麻愈伤组织诱导的最佳外植体是剑麻叶片,其出愈率最高,愈伤量最大。在整个诱导和分化过程中,剑麻对N的需求不大,而低铵盐含量对愈伤组织的诱导和分化有利;在含有较高浓度的盐酸硫胺素(VB1)的培养基中,愈伤组织的诱导率、愈伤量和分化率都有显著提高;Cu、Mo、Co等微量元素和有机成分肌醇,对剑麻愈伤组织的诱导并非必需,但对愈伤组织的分化可能是极有利的。The effects of four kinds of basic media and three kinds of sisal explants on the callus induction and differentiation of Agave sisalana were studied. The results showed that SH had the best effect on induction and differentiation of callus. The ratio of callus was almost 100%, the weight of callus was very large and the ratio of differentiation is 21.1%. Sisal leaves had the best effect on induction of callus with the best ratio and the largest weight of callus. Sisal had low demand for nitrogen throughout the process of callus induction and differentiation, and lower concentration of ammonium salt was advantageous to induction and differentiation of callus. Higher concentration of hydrochloric acid thiamine (vitamin BI) in the media increased callus induction and differentiation significantly. It seemed that Cu, Mo, Co and inositol were not essential to the induction of sisal callus, and they may be very favorable for callus differentiation.
