摘  要:采用ISSR标记对陕西核桃实生居群39个个体的遗传多样性水平及居群的遗传结构进行了研究,从13个引物中筛选出8个用于正式扩增,检测到48个位点为多态位点,多态位点百分率(PPB)为87.27%;居群材料间遗传相似系数GS变幅为0.473~0.836,平均达0.679;Shannon-Weaver遗传多样性指数的分布范围为1.483~1.960,Simpson指数的分布范围为0.739~0.846。UPGMA聚类分析结果具有一定的相似性,但是也存在明显差异。ISSR标记能将39份核桃种质完全区分开,为科学合理地保护和利用现有的核桃资源提供了理论依据和技术支持。The genetic diversity of 39 individuals and genetic structure of the population of Shanxi walnut ( Juglans regia L. ) were analyzed by inter - simple sequence repeat markers (ISSR). Eight primers were selected from 13 pre -screened primers for PCR. With them, 48 polymorphic loci were detected with the percentage of polymorphic bands (PPB) as 87.27%. The ISSR - derived genetic similarity (ISSR - GS) ranged from 0. 473 - 0. 836 with a mean of 0. 679. The Shannon - Weaver index of genetic diversity ranged from 1. 483 to 1. 960 and the Simpson index ranged from 0.739 to 0.846. The UPGMA cluster analysis showed certain similarity to it, but also presented obvious differences. In conclusion, the ISSR markers could separate the 39 individuals, which provided theoretical reference and technical support for the scientific protection and utilization of walnut resources.
