天舒胶囊效应部位组对硝酸甘油致大鼠偏头痛的影响

摘　　要：目的:研究天舒胶囊效应部位组对偏头痛动物模型的镇痛作用,并从神经生化代谢紊乱方面探讨其作用机制。方法:将健康SD大鼠随机分为正常对照组、模型组、西比灵组、天舒胶囊组、天舒胶囊效应部位3个剂量组,每组10只。皮下注射硝酸甘油(NTG)注射剂复制实验性偏头痛动物模型,观察注射硝酸甘油后60 min内大鼠行为反应,6 h后取血及脑组织检测一氧化氮(NO)、一氧化氮合酶(NOS)、β-内啡肽(β-EP)、降钙素基因相关肽(CGRP)。结果:天舒胶囊组、天舒胶囊效应部位高、中剂量组动物挠头次数显著减少,血浆和脑组织中NOS、NO含量显著降低,脑组织中β-EP含量显著升高,CGRP含量显著降低,使之始终趋于生理状态下的平衡水平。结论:天舒胶囊效应部位组具有缓解偏头痛症状,可通过有效调节NOS、NO、β-EP和CGRP的含量来达到治疗偏头痛的作用。This study was aimed to evaluate the analgesic effect of effective fractions of Tianshu capsule （TSC） on mi- graine animal model. Mechanism of effective fractions of TSC in the treatment of migraine was also investigated from the aspect of neurobiochemical metabolic disorders. Healthy SD rats were randomly divided into 4 groups, which were the normal control group, model group, sibelium group, TSC group and three dosage groups of effective fractions of TSC. Each group contained 10 rats. Experimental migraine animal model was duplicated by subcutaneous injection of nitroglycerin （NTG）. In the study, behavioral response of rats in 60 minutes was observed, and contents of NO, NOS, β-EP and calcitonin gene related peptide （CGRP） in blood and brain were detected in 6 hours. The results showed that in TSC group and the high-dosage and medium-dosage group of effective fractions of TSC, the animal head- scratching frequency was significantly decreased. The content of NOS and NO in blood and brain tissue were significantly reduced. The content of β-EP in brain tissues was obviously increased. And the content of CGRP was significantly decreased. This effect is able to maintain an equilibrium level. It was concluded that the effective fractions of TSC can relieve symptoms of migraine through its function of adjusting contents of NOS, NO,β-EP and CGRP.
