摘 要:目的:优选人参多糖的最佳除蛋白工艺。方法:以蛋白清除率、多糖保留率和糖醛酸保留率为指标,比较了两种不同除蛋白方法(Sevag法和TCA法)的效果,并采用正交试验优选最佳除蛋白工艺。结果:Sevag法的除蛋白效果优于TCA法,人参多糖的最佳除蛋白工艺为5%人参多糖溶液中加入1/3倍体积的Sevag试剂,V氯仿∶V正丁醇为4∶1,共除6次。结论:该工艺除蛋白效率较高,可用于人参多糖的分离精制。This study was aimed to optimize conditions for protein-removing from ginseng polysaecharides. The pro- tein-removing rate, retaining rate of polysaccharides and uronic acid were used as indicators. Results of two protein- removing methods (i.e., Sevag method and TCA method) were compared. And the optimized protein-removing condi- tions were obtained by orthogonal test. The results of Sevag method were better than TCA method. The optimized con- dition for protein-removing from ginseng polysaccharides was that Sevag reagent volume was equal to 1/3 times of 5% ginseng polysaccharides solution. And the volume ratio of CHC13 and normal butanol was 4:1. The protein-removing process was conducted 6 times. It was concluded that the method is efficient and can be used for the separation and refinement of ginseng polysaccharides.
