摘 要:在氧化应激状态下,转录因子Nrf2从细胞质转移入细胞核,与抗氧化反应元件结合调节多种保护性细胞因子表达,从而增强细胞对氧化应激的抵抗能力。研究表明,Nrf2通路在创伤性脑损伤(traumatic brain injury,TBI)后激活,抑制继发性脑损伤而发挥脑保护作用。而Nrf2基因敲除小鼠比野生型小鼠在TBI后炎性细胞因子表达增强,抗氧化和解毒酶活力下降,神经元钙超载、凋亡及脑水肿加重,神经功能缺失加重。以上研究提示,Nrf2通路在TBI中具有脑保护作用,另外,该通路还具有对TBI后继发性急性肺损伤、肠道屏障功能破坏的保护作用。Nrf2激活剂能减轻TBI后继发性脑损伤、维持血脑屏障功能、减轻脑水肿而发挥保护作用。In the oxidative state,transcription factor Nrf2 translocates from plasma to the nuclear and binds to the antioxidative response element,initiating expression of multiple genes to protect cell from oxidative stress.The Nrf2 pathway is activated after traumatic brain injury,which protects the brain from secondary injury.Comparing with the wild type counterpart,the Nrf2 knockout mice exhibited exacerbated inflammatory response,lower anti-oxidative and detoxicating enzyme activities,severer calcium overload in the neuron,deteriorated apoptosis and brain edema,and the neurological function was severely impaired following brain injury,these findings suggested that the Nrf2 pathway played a neuroprotective role in traumatic brain injury.At the same time,the Nrf2 pathway also played a protective effect against acute lung injury and gut barrier impairment secondary to traumatic brain injury.Activators of Nrf2 played a protective role by alleviating secondary brain injury,maintaining the blood-brain-barrier integrity and erase brain edema.
