摘　　要：目的探讨SD大鼠骨髓间充质干细胞的体外分离、传代培养方法,并对培养细胞进行鉴定。方法贴壁培养法分离SD大鼠骨髓细胞,传代扩增,倒置相差显微镜进行形态学观察,流式细胞术鉴定细胞。结果原代细胞较多细胞集落,细胞规则,呈梭形、椭圆形,传代细胞多成纤维形状,第3代培养细胞表面分子CD44,CD90,CD105呈阳性表达,但不表达CD34。结论贴壁培养法培养的大鼠骨髓原代细胞可稳定表达干细胞表面的标记分子。Objective To study the isolation,cultivation and preliminary identification in vitro of bone marrow mesenchymal stem cells（BMSCs） in SD rats.Methods The bone marrow cells in SD rats were separated and expanded by adherence culture,then subcultured and observed for morphology under invert microscope.The BMSCs were identified with flow cytometry.Results More primary cell colonies with regularly fusiform or oval cell were observed.The subcultured cells were mostly fibrous.At the 3rd passage,BMSCs showed high homogeneity and were positive for CD44,CD90 and CD105,and negative for CD34.Conclusion BMSCs in SD rats for isolation and cultivation by adherence culture could steadily express the stem cell surface markers.
