摘  要:目的探讨约氏疟原虫(P.yoelii17XL)Pys48核酸疫苗免疫BALB/c小鼠后免疫功能特点。方法将Pys48核酸疫苗肌肉内注射免疫BALB/c小鼠,并以空质粒注射组和正常小鼠作为对照,3次免疫后4w,处死小鼠;采用ELISA分别对疫苗免疫组和对照组小鼠脾细胞上清的IFN-γ、IL-4进行检测;流式细胞术检测脾中树突状细胞(Dendritic cells,DCs)亚群数量,表达TLR9的DCs的百分含量,以及分泌IgG抗体的B细胞百分含量。结果核酸疫苗免疫组IL-4水平较空质粒对照组和正常对照组均明显升高(P<0.05),IFN-γ水平与其它两组相比差异无统计学意义;脾中髓样DCs(mDCs)的数量在3组间无明显差异,浆细胞样DCs(pDCs)的数量在核酸疫苗免疫组出现有意义地升高,且表达TLR9的DCs的百分含量及分泌IgG抗体的B细胞百分含量均明显高于空质粒对照组和正常对照组(P<0.05)。结论 Pys48核酸疫苗具有良好的免疫原性,免疫小鼠后,通过增强特异性体液免疫发挥疫苗效应。To evaluate the characteristics of immune function after immunization by P. yoelii 17XL Pys48 DNA plasmid, the BALB/c mice were intramuscularly injected with Pys48 DNA plasmid and empty plasmid as control. The mice were sacri- ficed after the 3rd cycle of immunization. The levels of IFN-7, IL-4 in splenocyte supernatant obtained from each group!mice were assayed by ELISA. Flow cytometry was applied to quantitatively analyzed subset of DCs and DCs expressing TLR9, and B cells secreting IgG. It was found that IL-4 level from Pys48 DNA vaccine immunized mice increased obviously, compared to that from empty plasmid and normal control group (P〈0.05). And there was no obvious difference in IFN-7 level between each group. The number of splenocyte mDCs had no apparent disparity, while pDCs number from vaccine immunized nfic6 increased significantly. Moreover, the percentage contents of DCs expressing TLR9 and B cells secreting IgG were higher than those of other two control groups (P〈0.05). These results indicated that Pys48 DNA plasmid possessed a pretty immunogenicity and could develop vaccine effect through strengthening humoral immunity.
