Optimization of Culture Mediums for Stock Spawn and Liquid Strain of Armillaria mellea Symbiotic with Gastrodia elata

摘  要:通过正交试验获得一种蜜环菌母种培养基新配方(葡萄糖2.5%、酵母粉0.2%、乙醇0.5%、K2PO40.03%、MgSO40.015%、VB10.001%、琼脂粉1.5%),在菌丝体产量和性状方面优于目前常用的固体PDA加富培养基。利用板栗细小枝条和玉米芯的提取液添加至普通PDA培养基中,获得改良的蜜环菌液体原种培养配方(马铃薯8%、蔗糖0.8%、板栗细小枝段3%、玉米芯碎块3%)。与液体PDA加富培养基相比,菌球数目更多、个体更大、健壮度更高,且菌丝体产量最高可达2.5倍。板栗液玉米芯液组合使用后效果优于单独添加,以比例1:1效果最佳。A new medium (glucose 25%, yeast extract 2%, ethanol 0.5%, K2PO4 0.03%, MgSO4 0.015%, VB1 0.001% and agar powder 1.5%) for stock spawn of Armillaria mellea was obtained by orthogonal test. By using this medium, the growth of mycelia was superior to that of the widely used PDA solid medium both in biomass and traits. Another sub- merged culture medium (potato 8%, sucrose 0.8%, Castarea mollissima twigs 3%, chipped Corncob 3%) was also in- vented by adding Castarea mollissima twigs and corncob extracts into PDA medium. Compared to liquid enriched PDA medium, A. mellea in this medium can produce bigger, stronger mycelia and up to 2.5 times in biomass, and save 60% of potato and sugar use. Suitable proportions adding of Castarea mollissima or corncob extract could promote the growth of A. mellea, but Castarea mollissim extract is superior to corncob extract. The effect of combined two extracts is better than added separately, and the optimal ratio is 1:1.
