摘  要:目的研究兔颅脑爆炸伤后脑皮质中Caspase-3、细胞色素C(Cyt C)的表达情况。方法新西兰大白兔80只,随机分为对照组(n=10)和实验组(n=70)。实验组采用纸雷管制作兔颅脑爆炸伤模型,对照组不行爆炸伤。实验组根据处死时间再分为伤后1 h、6 h、12 h、24 h、3 d、7 d、14 d组,每组10只。采用免疫组化技术观察各组兔脑皮质Caspase-3、Cyt C的表达情况。结果 Caspase-3在实验组致伤后1 h出现明显表达,伤后24 h达高峰,随后开始下降,伤后7 d仍维持较高水平;伤后1 h^7 d与对照组比较,差异均有统计学意义(P<0.05)。Cyt C在实验组兔致伤后1 h即出现表达,伤后12 h达高峰,伤后24 h开始下降;伤后1~24 h与对照组比较,差异均有统计学意义(P<0.01)。结论兔颅脑爆炸伤后早期神经细胞胞质中Cty C增加可能是脑皮质细胞凋亡的关键步骤。Objective To investigate the expressions of Caspase-3 and cytochrome C(Cyt C) in cerebral cortex after brain explosive injury in rabbits.Methods Eighty New Zealand rabbits were randomly divided into control group(n=10) and experiment group(n=70).Brain explosive injury model was established by paper detonator in experiment group,and no explosive injury was performed in control group.After successful modeling,the experiment group was subdivided into 7 groups,namely groups 1 h,6 h,12 h,24 h,3 d,7 d,14 d after injury,according to sacrifice time point and each group had 10 rabbits.The expressions of Caspase-3 and Cyt C in cerebral cortex were observed by immunohistochemistry technique in every group.Results The expression of Caspase-3 in experiment group increased at 1 h after injury and reached peak at 24 h,and then decreased,and maintained high level on 7 d after injury.There was significant difference between 1 h to 7 d after injury in experimental group and control group(P0.05).The expression of Cyt C in experiment group increased at 1 h after injury and reached peak at 12 h,and decreased at 24 h.There was significant difference between 1 to 24 h after injury in experimental group and control group(P0.01).Conclusions The increased expression of Cyt C in cytoplasm of nerve cells may be the key step of apoptosis early after brain explosive injury in rabbits.
