摘 要:目的:探讨力竭运动对大鼠下丘脑室旁核内NF-κB、Bcl-2、Bax表达的影响。方法:SD大鼠随机分为对照组和力竭运动组,对照组10只,不进行游泳运动。力竭运动组30只,每天大鼠尾部负重(2%体重的重物)游泳1次至力竭,连续7 d,分别于最后一次运动后即刻(0 h)、4 h、8 h、12 h、16 h、24 h等不同时间点取材,采用Western blot技术检测大鼠下丘脑室旁核内NF-κB、Bcl-2、Bax蛋白含量,采用免疫组织化学染色分析NF-κB、Bcl-2、Bax免疫阳性神经元的定位和表达。结果:(1)Western blot结果:对照组与力竭运动0 h组NF-κB表达较低,力竭运动组运动后4 h NF-κB表达显著高于对照组(P<0.05),8 h达到峰值,而后下降,24 h时仍显著高于对照组(P<0.05)。力竭运动后即刻、4 h、8 h、12 h、16 h、24 h,Bcl-2表达均显著高于对照组(P<0.05),运动后12 h达到峰值。Bax表达与Bcl-2相似,力竭运动后即刻、4 h、8 h、12 h、16 h、24 h均显著高于对照组(P<0.05),运动后12 h达到峰值。(2)免疫组化染色结果:NF-κB与Bax阳性神经元数目表达与West-ern blot结果一致。对照组可见一定数量Bcl-2阳性神经元,力竭运动后Bcl-2阳性神经元数量有所增加,但各时间点之间比较差异无统计学意义(P>0.05)。结论:力竭游泳运动中下丘脑室旁核内NF-κB的活化可能参与中枢神经系统的细胞凋亡的诱发,表现为促凋亡蛋白Bax表达增高,而抑凋亡蛋白Bcl-2表达不明显。Objective To observe the expression of NF-κB,Bcl-2,and Bax in hypothalamus pavaventricular nuclei(PVN) of adult rats after exhausted exercise.Methods SD rats were divided into the control group and exercise group,rat exercise model was established through exhaustive swimming for a week.At 0 h,4 h,8 h,12 h,16 h,and 24 h after swimming,the cerebral tissues in PVN were sampled and the changes in NF-κB,Bcl-2,and Bax in PVN were detected by Western blot.The localization and expression of NF-κB,Bcl-2,and Bax immunopositive neurons were observed with immunohistochemical staining.Results(1)NF-κB expression both in control group and in exercise group 0h after exercise was lower,while 4h after exercise in exercise group was higher than control group(P 0.05) and reached its peak 8h after exercise.Expressions of Bcl-2 0 h,4 h,8 h,12 h,16 h,and 24 h after exercise were significantly higher than control group(P 0.05),and the peak expression appeared 12 h after exercise.The expression of Bax was similar to that of Bcl-2.(2)The numbers of NF-κB and Bax positive neurons detected by immunohistochemistry was the same as by Western blot.There were some Bcl-2 positive neurons in the control group without statistical difference(P 0.05) from the exercise group.Conclusion The activation of NF-κB in PVN after exhaustive exercise probably involved in the apoptosis of CNS.
