摘  要:目的研究嗅鞘细胞移植对大鼠损伤脊髓内的神经生长因子(NGF)表达的影响,以从NGF角度探讨嗅鞘细胞移植修复大鼠脊髓损伤的机制。方法48只SD大鼠用NYU-Ⅱ撞击机(10g-25mm)损伤T10脊髓制作脊髓损伤(SCI)模型,随机分为嗅鞘细胞组、DMEM组各24只;另设立正常对照组6只。将GFP-嗅鞘细胞细胞悬液移植人嗅鞘细胞组大鼠损伤处,DMEM组用单纯的DMEM/F12液代替,正常对照组不做任何处理。移植术后1d、7d、14d、21d,用BBB评分法测定脊髓运动功能,RT—PCR方法比较各时间点NGF表达差异。移植术后第21天应用免疫组化比较嗅鞘细胞组、DMEM组、正常对照组大鼠脊髓损伤区NGF的表达差异。结果移植后1d、7d、14d、21d,嗅鞘细胞移植组运动功能评分均高于同期DMEM组。NGF表达量在术后第1天最高,第7天达峰值,之后缓慢降低。移植后第21天,嗅鞘细胞移植组脊髓NGF表达量高于同期DMEM组和正常对照组。结论嗅鞘细胞移植可上调损伤脊髓NGF的表达,从而促进了损伤脊髓的修复。Objective In order to approach the mechanism of the spinal cord injury (SCI) repaired by OECs transplantation, the effects of olfactory ensheathing cells (OECs) transplantation on the expression of the nerve growth factor (NGF) in injured spinal cord was quantitatively studied. Methods The rat models of SCI were made by NTY - II impactor ( 10g -25 mm) at T10 level. There are 6 rats in the normal group and 48 rats in the injuried group. Then the injuried group was randomly divided into OECs transplantation group and DMEM group. The GFP- OECs were transplanted into the injured region immediately after SCI in the OECs transplantation group. The DMEM group used the DMEM/F12 to take the place of GFP - OECs. Nothing to do with normal control. Then the locomotor function was appraised by BBB score at 1 day, 7 day, 14 day and 21 day. The expression of the NGF in the injured region was detected by the methods of immunohistocbemistry and RT - PCR. Results The BBB score in OECs transplantation group was higher than the DMEM group and normal group at 1 day, 7 day, 14 day and 21 day. The expression of the NGF was advanced at 1 day, and highest at 7 day. The expression of the NGF in the OECs group was higher than the normal control group and the DMEM group at 21 day. Condusions The expression of NGF can be up - regulated by OECs transplantation, which may be one of the mechanisms for OEC's repairing injured spinal cord.
