摘  要:目的探讨油樟叶挥发油及其主要组成成分(1,8-桉叶油素、黄樟油素、γ-松油烯、α-松油烯和松油烯-4-醇)的抗肝癌活性。方法运用体外培养细胞四甲基偶氮唑盐(MTT)快速比色法,观察油樟叶挥发油的主要组成成分(1,8-桉叶油素、黄樟油素、α-松油烯、松油烯-4-醇和γ-松油烯)对人肝癌BEL-7402细胞的抑制作用,并用软琼脂细胞集落形成实验,观察油樟叶挥发油的主要组成成分对人肝癌BEL-7402细胞形成细胞集落的抑制作用。结果油樟叶挥发油、1,8-桉叶油素、黄樟油素、γ-松油烯、α-松油烯和松油烯-4-醇体外对人肝癌BEL-7402细胞的最大增殖抑制率分别为58.63%、42.83%、90.04%、81.53%、34.83%和31.46%;对人肝癌BEL-7402细胞集落形成的最大抑制率分别为55.13%、61.99%、94.56%、89.54%、62.02%和59.91%。结论油樟叶挥发油及其主要组成成分能不同程度地抑制人肝癌BEL-7402细胞的体外增殖及单个细胞形成细胞集落。其中黄樟油素和γ-松油烯对人肝癌BEL-7402细胞的抑制作用最强。Objective In order to study the anticancer activity of essential oil from Cinnamomum longepaniculatum leaves and its major components ( 1,8-cineole, sassafras oil, 3,-terpinene, a-terpinene and terpinene-4-alcohol) against human BEL-7402 ceells. Methods By 3-4,5-diemethylthiarol-2,5-diphenytetrazolium bromide (MTT) fast staining method to study the growth inhibition rate of the essential oil from Cinnamomum longepaniculatum leaves and its major components on human BEL- T402 cells, and by soft agar cloning formation assay to study the effect of the essential oil from Cinnamomum longepaniculatum leaves and its major components against human BEL-7402 cells. Results The growth inhibition rates of the essential oil from Cinnamomum longepaniculatum leaves, 1,8-cineole, sassafras oil, a-terpinene, a-terpinene and terpinene-4-alcohol against the BEL-7402 cells were 58. 63% , 42. 83% , 90. 04% , 81.53%, 34. 83% and 31.46% , respectively in vitro. Colony forming rate of the essential oil from Cinnamomum longepaniculatum leaves, 1,8-cineole, sassafras oil, γ-terpinene. a-terpinene and terpinen- 4-alcohol on BEL-7402 ceils were 55. 13%, 61.99%, 94. 56%, 89. 54%, 62. 02% and 59. 91%, respectively. Conclusion The essential oil from Cinnamomum longepaniculatum leaves and its major components can inhibit the proliferation of liver cancer cell Bel-7402 and reduce the forming cell colony. Sassafras oil and γ-terpinene have exceptional inhibitory ability.
