肠安I号及其低浓度原药安慰剂对腹泻型IBS大鼠血清TNF-α、IL-10的影响

摘　　要：目的探讨肠安Ⅰ号及含有低浓度肠安Ⅰ号成分的安慰剂对腹泻型肠易激综合征大鼠血清细胞因子TNF-α、IL-10的影响。方法大鼠随机分为正常组,模型组,含5%、含10%肠安Ⅰ号组,肠安Ⅰ号高、中、低剂量组,得舒特组共8组,每组10只。采用番泻叶灌胃+传统束缚应激刺激复制腹泻型肠易激综合征大鼠模型。各组给予相应的干预措施。治疗结束后,测定大鼠的腹泻指数和血清TNF-α、IL-10的含量。结果与正常组比较,模型组各指标差异有统计学意义(P<0.05,P<0.01),说明造模成功;与模型组比较,肠安Ⅰ号各剂量组血清中TNF-α、IL-10水平、腹泻指数差异有统计学意义(P<0.05,P<0.01);与模型组比较,含5%、含10%肠安Ⅰ号组各指标差异无统计学意义(P>0.05)。结论肠安Ⅰ号高、中、低剂量可通过增加IL-10的表达,降低TNF-α的含量,调节促炎因子\抗炎因子水平,使其抗炎活性增强,从而发挥对ⅠBS的治疗作用;而含5%、含10%肠安Ⅰ号加辅料组没有药理作用,仅从中药的色、味方面模拟了肠安Ⅰ号的特点,起到了安慰剂对照作用。Objective To investigate the effect of ＂Chang＇an Ⅰ＂ and placebo on TNF-α and IL-10 in rats with irritable bowel syndrome（IBS） of diarrhea pattern.Methods The rats were randomized into 8 groups： normal group,model group,5% ＂Chang＇an Ⅰ＂ group,10% ＂Chang＇an Ⅰ＂ group,＂Chang＇an Ⅰ＂ groups（high,medium and low dose） and Dicetel group,10 rats in each group.The models with IBS were duplicated by intragastric administration with senna leaf and traditional restraint stress,and different treatments were taken for different groups.After treatment,the diarrhea index,and the content of TNF-α and IL-10 were detected.Results There was significant difference in all the indicators between normal group and model group（P0.05,P0.01）,it showed the models were established successfully.There were significant differences in TNF-α,IL-10 and diarrhea index between model group and ＂Chang＇an Ⅰ＂ groups（P0.05,P0.01）;no significant difference was found in all indicators among model group,5% ＂Chang＇an I＂ group and 10% ＂Chang＇an I＂ group（P0.05）.Conclusion ＂Chang＇an Ⅰ＂ groups（high,medium and low dose） can treat IBS by improving expression of IL-10 to decreasing the content of TNF-α,regulating the levels of pro/anti-inflammatory cytokines to enhance the anti-inflammatory activities;while the 5% ＂Chang＇an Ⅰ＂ and 10% ＂Chang＇an I＂ group imitated the therapeutic effect of ＂Chang＇an Ⅰ＂ groups from the color and taste,can be a placebo without any pharmacological action.
