摘  要:目的研究芒柄花黄素对脑缺血再灌注损伤小鼠炎症反应的影响。方法将50只昆明种小鼠随机分为脑缺血再灌注组,芒柄花黄素高、中、低剂量组,假手术组。给药组术前连续灌胃7d,于末次给药60min后行手术,夹闭双侧颈总动脉后松开,造成全脑缺血再灌注模型,观察缺血30min再灌注72h后,芒柄花黄素对各组小鼠脑含水量、脑组织中TNF-α、IL-β的影响。结果与脑缺血再灌注模型组相比,芒柄花黄素能够减轻小鼠的脑缺血再灌注损伤程度,降低脑含水量,降低脑组织中的IL-1β、TNF-α含量。结论芒柄花黄素对小鼠全脑缺血再灌注损伤具有保护作用,其机制可能与其影响脑缺血再灌注后的炎症反应有关。Objective To investigate the effect of formononetin on inflammatory response induced by cerebral ischemia/reperfusion(I/R) injury in mice.Methods Fifty Kunming mice were randomly divided into I/R group,sham-operated group,high-,medium-and low-dose of formononetin groups.The animals were intragastrically administered with drugs for 14 days.The operation was performed to induce the ischemia/reperfusion injury model in mice.After ischemia for 30 mins and repefusion for 72 hours,the influences of formononetin on the brain water content,the levels of TNF-α and IL-1β in brain tissue were measured.Results Compared with I/R group,formononetin could obviously reduce the brain water content,the levels of TNF-α and IL-1β in the brain tissue(P0.01 or P0.05).Conclusion Formononetin has protective effect on cerebral ischemia/reperfusion injury,which the mechanism may be related to decreasing inflammatory response.
