摘 要:本研究用混合酶法分离培养人脐带间充质干细胞(MSC),检测在不同浓度17β-雌二醇(E2)作用下MSC中MicroR-NA的表达情况。进一步通过转染使MSC高表达miR-146a,并分析转染后MSC的表型、功能及相关因子的表达变化。结果显示,E2处理的MSC中IFN-β的表达没有发生明显变化,STAT1与IDO1的表达显著下调。同时,E2处理能够上调MSC对miR-146a的表达,降低其对miR-23b和miR-21的表达。miR-146a转染不影响MSC的表型,而显著降低STAT1和IDO的表达。提示E2可能通过影响MSC内miR-146a的表达改变其免疫调节功能。We used the mixed enzyme method for the isolation and culture of human umbilical cord mesenchymal stem cells(MSC),and investigated the expression of MicroRNA in MSC treated with 17β-Estradiol(E2) at different concentrations.Then the phenotypes,functions and the expression of related genes of MSC were analyzed after transfecting miR-146a into MSC.Our results showed that the expression of IFN-β did not change significantly in MSC treated with E2,while the expression of STAT1 and IDO1 were remarkably downregulated.The expression of miR-146a was remarkably enhanced while the expression of miR-23b and miR-21 were decreased.The transfection of miR-146a significantly inhibited the expression of STAT1 and IDO,without affecting the phenotypes of MSC.Therefore,we conclude that E2 may alter the immunoregulatory effects of MSC by modulating miR-146a expression.
