注射性坐骨神经损伤后针刺腧穴对家兔胫骨前肌胶原纤维和NADH-TR的影响

摘　　要：目的探索注射性坐骨神经损伤后针刺腧穴对家兔胫骨前肌内胶原纤维和NADH-TR的影响。方法成年家兔35只,分正常、青霉素注射、生理盐水注射、青霉素注射加针刺腧穴治疗4组;手术暴露坐骨神经干,神经外膜下注射青霉素或生理盐水0.5 mL;青霉素注射损伤4周后针刺患侧环跳和后三里4～8周;VG染色法和NADH-TR组化染色法观察胶原纤维与NADH-TR的变化。结果青霉素注射坐骨神经后2周,胫骨前肌内胶原纤维已明显增生;NADH-TR活性增强,三型肌纤维构成比发生了变化,肌纤维横切面积减小,光密度值下降,肌重减轻;4周时进一步加重,肌块明显萎缩。生理盐水注射2周,可见肌内一过性损伤。青霉素注射4周后针刺腧穴治疗4周,上述指标明显好转,治疗8周时进一步恢复,接近正常。结论针刺腧穴治疗可减少肌内胶原纤维增生,降低NADH-TR活性,改善肌萎缩。Objective To explore effecting of collagen fiber and NADH-TR of tibialis anterior of rabbit by sciatic nerve injection injury and acupuncture treatment.Methods 35 adult healthy rabbit were used,they were randomized into four group：ie the normal,penicillin injection,normal saline injection,penicillin injection and acupuncture treatment respectively.Sciatic nerve-trunk were exposed through surgery and penicillin or normal saline 0.5ml were injected under the perilemma.After penicillin injection injury 4 weeks,the trouble Foot Three Li and Circular Jump were acupunctured 4-8 weeks,then VG and NADH-TR staining method was adopted to observe the changes of collagen fibers and NADH-TR in the rabbit＇s tibialis.Results After penicillin injecting into sciatic nerve-trunk perilemma 2 weeks,the collagen fibers hyperplasied obviously in tibialis anterior muscle,NADH-TR activation strengthened,the constituent ratio of three muscle fiber types have changed;crosscut area and optical density of muscle fiber and weight of muscle was dropped;they made more serious at fourth week,muscular atrophy have been observed obviously.while saline injection caused only transient impairment during 2 weeks in the muscle.If two acupoint were acupunctured 4 weeks after penicillin injection injury 4 weeks,above-mentioned index wloud take a favorable turn markedly,and to treat 8 weeks,moved forward a single step recovery to near normal.Conclusions Acupuncture treatment could reduce collagen fibers hyperplasia,degrade NADH-TR activation,improve amyotrophy in tibialis anterior of rabbit.
