Optimization of Ultrasonic Extraction Technology of Total Alkaloid from Fritillaria thunbergii by Response Surface Methodology

摘  要:目的:优选浙贝母中总生物碱的超声波提取工艺。方法:选取提取时间、超声波功率和液料比为随机因子,在单因素试验的基础上,进行3因素3水平的Box-Behnken试验设计,以总生物碱得率为响应值进行分析。结果:最佳提取条件为提取时间48min、超声波功率420W、液料比6.6(mL:g)。在此条件下,总生物碱得率验证值为0.1685%。结论:响应面法优选浙贝母中总生物碱的超声波提取工艺简便、易行。OBJECTIVE: To optimize the ultrasonic extraction technology of total alkaloid from Fritillaria thunbergii. METHODS: Extracting time, ultrasonic power and liquid-solid ratio were selected as casual factors, based on single-factor experiments, three-factor-three-level Box-Behnken experiment design method was applied, and the extraction rates of total alkaloid were used as response values for response surface methodology (RSM) analysis. RESULTS: The optimum conditions of ultrasonic extraction were as follows: extraction time of 48 min, ultrasonic power of 420 W, and liquid-solid ratio of 6.6 (mL:g). Under these conditions, the extraction rate of total alkaloid was 0.168 5%. CONCLUSION: The extraction process optimized by RSM analysis is simple and more convenient for extracting total alkaloid from E thunbergii.
