口服戊酸雌二醇对自然更年期大鼠空腹血糖、血脂及胰岛素抵抗的影响

摘　　要：目的探讨口服戊酸雌二醇对自然更年期雌性Sprague-Dawley(SD)大鼠空腹血糖(FBG)、血脂及胰岛素抵抗的影响。方法将70只15月龄雌性更年期SD大鼠随机分为实验组和对照组,每组35只。实验组每天给予0.21 mg.kg-1戊酸雌二醇灌胃给药,对照组给予等量生理盐水灌胃,14周后腹主动脉采血,测定血清雌二醇(E2)、卵泡刺激素(FSH)、FBG、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、总三酰甘油(TG)、胰岛素抵抗指数(HOMA-IR)和胰岛素敏感指数(HOMA-ISI)。结果实验组SD雌性大鼠在戊酸雌二醇灌胃14周后血清E2水平和HOMA-ISI明显高于对照组(P<0.05),血清FSH、FBG、TC、TG、LDL-C和HOMA-IR均明显低于对照组,差异有统计学意义(P<0.05),HDL-C较对照组有升高趋势,但差异无统计学意义(P>0.05)。结论口服戊酸雌二醇可降低FBG,改善血脂,降低TC、TG、LDL-C,增加胰岛素敏感性。Objective To investigate the effects of oral estrogen on the fasting blood glucose（FBG）,lipids and insulin resistance in climacteric female Sprague-Dawley（SD） rats. Methods Seventy specific pathogen-free female SD rats,fifteen-month-old,were randomly divided into treatment group and control group,thirty-five in each group.The rats in treatment group were given estradiol valerate intragastric administration orally,once per day;the rats in control group were given the physiological saline intragastric administration,once per day,the two groups all were treated for fourteen weeks.After the treatment,animals,blood samples were taken to assess the level of estradiol（E2）,follicle stimulating hormone（FSH）,FBG,high density lipoprotein cholesterol（HDL-C）,low density lipoprotein cholesterol（LDL-C）,totalcholesterol（TC）,triglyeride（TG）,insulin resistance index（HOMA-IR） and insulin sensitivity index（HOMA-ISI）. Results Fourteen weeks after intragastric administration,in the treatment group,serum E2 level and HOMA-ISI were higher than those in control group（P0.05）;the levels of FSH,FBG,TC,TG,LDL-C and HOMA-IR were lower than those in control group,there were significant statistics differences（P0.05）;the level of HDL-C was higher than that in control group,but there was non-statistics difference（P0.05）. Conclusion Estradiol valerate by intragastric administration can decrease the FBG,improve the blood lipids,decrease the TC,TG and LDL-C level,and it can also increase insulin sensitivity.
