错配修复基因1在新疆维吾尔族及汉族卵巢子宫内膜异位症患者中的表达与意义

摘　　要：目的探讨新疆维吾尔族、汉族人群中错配修复基因1(hMLH1)转录水平与卵巢子宫内膜异位症发病的关系。方法收集新疆地区维吾尔族、汉族卵巢子宫内膜异位症患者异位内膜组织(异位内膜组)、在位内膜组织(在位内膜组)以及正常子宫内膜组织(对照组),在位内膜组及对照组的子宫内膜均通过病理检查分为分泌期或增殖期。采用实时荧光定量聚合酶链式反应(QRT-PCR)定量分析各内膜组织中hMLH1的表达水平。结果维吾尔族及汉族在位内膜组、对照组子宫内膜分泌期与增殖期hMLH1的表达量均无明显差异(P=0.807,P=0.956,P=0.511,P=0.286);维吾尔族和汉族异位内膜组与在位内膜组中hMLH1的表达量比较差异无统计学意义(P=0.058,P=0.854);维吾尔族在位内膜组及对照组的内膜组织中hMLH1的表达量中位数(M)分别为0.109、0.120,四分位数间距(Q)分别为0.180、0.306;汉族在位内膜组及对照组的内膜组织中hMLH1的表达量M分别为0.209、0.270,Q分别为0.286、0.309;hMLH1的表达在在位内膜组及对照组间比较差异有统计学意义(P=0.017),hMLH1的表达在维吾尔族、汉族间比较差异有统计学意义(P=0.037)。结论在位内膜组织中hMLH1的表达水平下降可能与新疆地区维吾尔族及汉族卵巢子宫内膜异位症发病有关。子宫内膜组织中hMLH1的表达水平存在民族差异。Objective To investigate the expression levels and the relationship with ovarian endometriosis pathogenesis of homo sapiens mismatch repair protein homolog 1（hMLH1） in Xinjiang Uygur and Han population. Methods Ectopic endometrial tissue（ectopic endometrium group）,eutopic endometrial tissue（eutopic endometrium group） and normal endometrial tissue（normal control group） of Uygur,Han patients with ovarian endometriosis in Xinjiang were collected.The endometrium of the eutopic endometrium group and the control group was divided into the secretory phase or the proliferation phase by pathological examination.Real-time fluorescence quantitative polymerase chain reaction（QRT-PCR） method was used to analyse the hMLH1 gene expression of various endometrial tissue. Results The hMLH1 expression levels in secretion phase and proliferation phase of endometrial showed no difference in endometrium group and control group in Uygur and Han people（P=0.807,P=0.956,P=0.511,P=0.286）;the expression of hMLH1 in ectopic endometrium group and eutopic endometrium group has no statistical significance in Uygur and Han people（P=0.058,P=0.854）;Median（M）,Quartile（Q） of hMLH1 expression both in eutopic endometrium group and control group of Uygur people were 0.109,0.120 and 0.180,0.306 respectively.M,Q of hMLH1 expression both in eutopic endometrium group and control group of Han people were 0.209,0.270 and 0.286,0.309 respectively.hMLH1 expression showed difference between eutopic endometrium group and control group（P=0.017）.There was difference in hMLH1 expression between Han and Uygur people（P=0.037）. Conclusion The decreasing hMLH1 expression in eutopic endometrium may be related with ovarian endometriosis of Uygur and Han people.And there is ethnic difference in hMLH1 expression of endometrium.
