HPLC法测定玉叶清火胶囊中穿心莲内酯和脱水穿心莲内酯的含量

摘　　要：目的:建立用反相高效液相色谱法测定玉叶清火胶囊中穿心莲内酯和脱水穿心莲内酯含量的方法。方法:采用Hyper-silODS2柱(C18,4.6×200mm,5μm),以流动相为甲醇-水(55∶45),流速为0.85ml.min-1,检测波长为250nm,进样量为10μl;柱温为室温。结果:穿心莲内酯和脱水穿心莲内酯分别在0.02698～0.2698mg·ml-1(r=0.9997)和0.02242～0.2242 mg·ml-1(r=0.9997)范围内,进样浓度与峰面积呈良好线性关系,穿心莲内酯和脱水穿心莲内酯的平均回收率分别为101.44%和103.94%,RSD为1.36%(n=5)和1.82%(n=5)。结论:该方法简便、准确,可作为玉叶清火胶囊质量控制的方法。Objective： To establish method for determining the content of andrographolide and dehydroandrographolide in Yuyeqinghuo Capsule by RP- HPLC. Methods： The HypersilODS2 （C18, 4. 6 × 200mm, 5μm） was used and mobile phase was methanolwater （55：45 ）, and its temperature was used at room temperature, with the flow rate of 0. 85ml · min- 1 and the wave- length of detection of 250 nm, and the injection amount of 10μl. Results： The linear ranges of andrographolide and dehydroandrographolide were at 0. 02698~0. 2698mg·ml-1（r=0. 9997）and0. 02242~0. 2242mg· m1-1 （r=0. 9997）, the averagerecoveries （n=5） were 101. 44% and 103. 94% with RSD 1. 36%, 1. 82%. Conclusions： The method was simple, accurate, and can be used for the quality control of Yuyeqinghuo Capsule.
