摘  要:目的探讨骨形态发生蛋白-2(BMP-2)、梗死心肌裂解液模拟的生物微环境诱导骨髓间充质干细胞(BMSCs)向心肌样细胞分化及在模拟的微环境中BMP-2对诱导的影响。方法 BMSCs分离与体外培养,构建大鼠心肌梗死模型制成梗死心肌裂解液;分4组:单纯DMEM培养组(A组)、梗死心肌裂解液组(B组)、梗死心肌裂解液加BMP-2阻断剂noggin组(C组)、BMP-2组(D组),通过倒置相差显微镜形态学观察、应用免疫细胞化学技术检测心肌特异性肌钙蛋白(cTnT)、心肌特异性肌球蛋白重链(MHC)的表达,并对诱导的心肌样细胞进行统计学分析。结果诱导3周后免疫细胞化学染色分析cTnT、MHC,A、C组呈阴性表达,B、D组呈阳性表达,B、D组与A、C组相比阳性细胞数增加(P<0.05)。结论 BMP-2、梗死心肌裂解液可诱导BMSCs分化为心肌样细胞;且BMP-2是梗死心肌裂解液诱导过程中重要的组成成分之一。Objective To investigate the influence of bone morphogenetic protein 2(BMP-2),infarcted myocardium lysate simulated biological microenvironment on bone marrow mesenchymal stem cells(BMSCs) differentiation into cardiomyocytes and the effect of BMP-2 on the induction in the simulation microenvironment.Methods BMSCs were separated and cultured in vitro.The rat model of myocardial infarction was constructed to prepare myocardial infarction lysate,which was divided into 4 groups:pure DMEM culture group(A),infarcted myocardium lysate group(B),infarcted myocardium lysate + BMP-2 blocker noggin group(C) and BMP-2 group(D).The morphological observation was performed by the inverted phase contrast microscope,the immunohistochemical technique was used for detecting the expression of cardiac specific troponin(cTnT) and myocardial specificity myosin heavy chain(MHC) expression,and the induced cardiomyocytes were analyzed by statistics.Results After 3-week induction,the immunocytochemistry analysis showed the negative expression of cTnT and MHC in the group A and C,and the positive expression in the group B and D.Comparing the group B and D with the group A and C,positive cells number was increased with statistical difference between them(P〈0.05).Conclusion BMP-2 and infarcted myocardium lysate can induce the differentiation of BMSCs into cardiomyocytes and BMP-2 is one of important components during infarcted myocardium lysate induction process.
