阿仑膦酸钠对血管钙化干预的研究

摘　　要：目的:观察阿仑膦酸钠对钙化的大鼠主动脉组织中骨桥蛋白(OPN)表达的影响,并探讨相关机制。方法:通过给实验大鼠注射维生素D3和华法林建立主动脉钙化模型(钙化模型组),给予模型大鼠阿仑膦酸钠干预治疗(阿仑膦酸钠组),并设正常对照组。采用Von Kossa染色观察各组主动脉的钙化沉积;采用RT-PCR检测各组主动脉OPN mRNA表达;采用免疫组织化学染色检测OPN蛋白表达;采用SpectrAA-240FS原子吸收分光光度计测定各组主动脉钙含量。结果:Von Kossa染色,正常对照组未见黑色深染结构,钙化模型组主动脉中膜有片状黑色深染结构,阿仑膦酸钠组主动脉黑色深染结构明显减少。钙化模型组大鼠主动脉钙含量比正常对照组显著升高(P<0.05),OPN mRNA和蛋白表达明显降低(P<0.05)。阿仑膦酸钠组主动脉钙含量比钙化模型组明显降低(P<0.05),OPN mRNA和蛋白表达显著升高(P<0.05)。结论:阿仑膦酸钠可能通过上调OPN表达来抑制大鼠主动脉血管钙化。Objective：To observe the effect of alendronate on osteopontin（OPN）in calcified aorta tissue of the rat,and explore the mechanism of vascular calcification.Method：An animal calcification model was established with vitamin D3 and warfarin（named calcification group）.Alendronate group was treated with vitamin D3 and warfarin plus alendronate,The control group was given daily subcutaneous injection of normal saline.There were 20 rats in each group.Mineral deposition was assessed by Von Kossa staining.OPN mRNA and protein expression were measured by RT-PCR and immunohistochemical staining respectively.Calcium content was determined by using an atomic absorption spectrophotometer（SpectrAA-240FS）.Results：Von Kossa staining showed no calcification can be detected in control group.Positively-stained black areas were observed in calcification group,and there was less calcification in alendronate group.Compared with control group,the aortic calcium content of calcification group was obviously increased,OPN mRNA and protein expression were reduced.Compared with calcification group,the aortic calcium content of alendronate group was obviously reduced,OPN mRNA and OPN protein expression were increased.Conclusion：Tunica media calcification of the rat aorta may be inhibited by alendronate by up-regulating OPN expression.
