摘 要:目的:观察噻唑烷二酮类药物对糖尿病大鼠肝脏chemerin、cmklr1及肿瘤坏死因子(TNF)-αmRNA表达的影响。方法:雄性SD大鼠随机分为正常组、糖尿病组与吡格列酮组,尾静脉注射链脲佐菌素制作糖尿病大鼠模型,造模成功后吡格列酮组每天按15mg/kg经胃灌药,连续给药8周。第8周末处死大鼠留取肝脏组织。采用实时定量PCR法检测大鼠肝脏chemerin、cmklr1及TNF-αmRNA的表达水平。结果:糖尿病组肝脏chemerin表达低于正常组及吡格列酮组,差异均有统计学意义(均P<0.017)。3组间肝脏cmklr1表达差异无统计学意义。糖尿病组肝脏TNF-α表达较正常组升高(P<0.017),吡格列酮组较糖尿病组有所降低,但差异无统计学意义。结论:糖尿病大鼠肝脏TNF-α表达增高,吡格列酮可能通过上调chemerin的表达而改善肝脏的胰岛素敏感性。Objective: To investigate the effect of thiazolidinediones on expression of chemerin, cmklrland tumor necrosis factor (TNF)-alpha mRNA in liver of diabetic rat model. Methods: Male SD rats were randomized into three groups, normal con- trol group, diabetes group and pioglitazone group. Murine diabetic models were induced with streptozotocin (STZ). After success- ful modeling, pioglitazone [15 mg/(kg, d)] was given to rats of pioglitazone group for 8 weeks. Rats were sacrificed to get liver tis- sues at the end of 8 weeks. Expressions of chemerin, cmklrl and TNF-α mRNA were detected by the method of real-time quanti- tative PCR. Results: Results of real-time quantitative PCR showed that expression of chemerin mRNA was significantly lower in diabetes group than that of normal control group and pioglitazone group (P 〈 0.017). NO significant difference was found in ex- pression of cmklrl mRNA between three groups. The expression of TNF-α mRNA was significantly higher in diabetes group than that of normal control group (P 〈 0.017). Conclusion: The expression of TNF-α mRNA was upregulated in the liver tissue of diabetes group. Pioglitazone may improve insulin sensitivity of liver by upregulating expression of chemerin mRNA.
