聚驱后油藏产多糖微生物多样性与功能分析

摘  要:通过16SrDNA克隆建库方法对大庆北二西油藏微生物调剖过程中和调剖后4个不同油井(B2-D4-P45井、B2-4-P47井、B2-4-P49井和B2-5-51井)3个不同时间段的样品经选择富集后的产多糖微生物群落进行研究。结果表明,主要的产多糖菌为爱媛类芽孢杆菌(Paenibacillus ehimensis)和爱媛芽孢杆菌(Bacillus ehimensis),经分离纯化得到一株纯菌DT-1,与爱媛类芽孢杆菌的16SrDNA序列同源性达到99%。调剖过程中主要激活了3类产多糖菌,调剖1年后微生物多样性明显增加,产生物表活剂菌和降解石油烃菌都被共同激活。16S rDNA clone library technique was used to analyze relative abundances of compositions of ploysaccharide bacterial profile control in the enrichment cultures of produced water in 3 different intervals from 4 different wells(such as Well B2-D4-P45,Well B2-4-P47,Well B2-4-P49 and B2-5-51) in the west of Region 2 in the north of Daqing Oilfield after polymer flooding.Results show that the dominant ploysaccharide bacteria are Paenibacillus ehimensis.A strain of DT-1 is isolated,phylogenetic analysis based on 16S rDNA gene sequence indicates that strain DT-1 is affiliated with Paenibacillus ehimensis with a similarity of 99%.During the process of the microbial plugging,three types of ploysaccharide bacteria are stimulated.One year after profile control,microbial diversities increase significantly,and biosurfactant-producing bacteria and petroleum hydrocarbon-degrading bacteria are co-activated.
