摘 要:目的观察中药复方冠心康对载脂蛋白E(ApoE)基因敲除(ApoE-/-)动脉粥样硬化小鼠主动脉粥样斑块病理形态及病灶处单核细胞趋化蛋白l(MCP-1)、核因子-κB(NF-κB)的影响。方法 50只6周龄ApoE-/-小鼠予高脂饲料喂养建立实验性动脉粥样硬化小鼠模型。10只6周龄C57BL/6J小鼠为正常组,给予普通饲料。至第12周龄时,将ApoE-/-小鼠随机分为模型组、辛伐他汀组及冠心康高(3.456 g/mL)、中(1.728 g/mL)、低剂量组(0.864 g/mL),继续给予高脂饲料,每日灌胃给药1次。连续灌胃8周后,取主动脉进行苏木素-伊红染色观察病理形态改变,免疫组织化学法检测斑块处MCP-1、NF-κB的表达含量。结果与模型组比较,各给药组管腔面积(LA)大、血管内膜厚度(IMT)薄、斑块面积(PA)小、纤维肌性成分(FS)少、脂质中心面积(CA)小、最小纤维帽厚度(FCT)薄、斑块面积与管腔面积之比(PA/LA)小、脂质中心面积与斑块面积之比(CA/PA)小、脂质中心面积与纤维肌性成分之比(CA/FS)大,MCP-1和NF-κB平均光密度值小(P<0.05)。冠心康高剂量组作用明显优于辛伐他汀组(P<0.05)。冠心康低、中、高剂量组之间比较不存在量效关系,但以冠心康高剂量组作用最为明显。结论冠心康对ApoE-/-小鼠动脉粥样硬化有一定的干预作用,可减少MCP-1、NF-κB的表达,延缓动脉粥样硬化的进展。Objective To observe the effects of Guanxinkang on the pathological morphology of atheromatous plaque and the expression of MCP-1,NF-?B of ApoE-deficient mice with atherosclerosis.Methods Fifty 6-week-old ApoE-deficient mice received a high-cholesterol diet were induced atherosclerosis and randomly divided into 5 groups:model group,simvastatin group and low-dosage(0.864 g/mL),medium-dosage(1.728 g/mL),and high-dosage(3.456 g/mL) Guanxinkang group.Another ten 6-week-old C57BL/6J mice were used as a normal control.atheromatous plaque in mouse's aortic was observed.After gavage of corresponding drugs once daily for 8 weeks continuously,by hematoxylin-eosin staining,and the expression level of MCP-1,NF-?B in the plaques was detected.Results Compared with the model group,after treatment,the luminal area(LA) and CA/FS were bigger,intima medium thickness and fiber cap thickness were thinner;plaque area(PA),fiber structure(FS),cholesterol area(CA),PA/LA,CA/PA were smaller.Meanwhile,the average optical density of MCP-1 and NF-?B were smaller.The effect of the high-dosage Guanxinkang group was obviously better than simvastatin(P0.05).There was no dose-effect relationship among the three dosage of Guanxinkang group,but the high-dosage Guanxinkang group was the best.Conclusion Guanxinkang can reduce the expression of MCP-1 and NF-?B,and delay the progression of atherosclerosis.
