参知健脑胶囊对东莨菪碱致记忆障碍小鼠中枢胆碱能系统的影响

摘　　要：目的观察参知健脑胶囊对获得性记忆障碍小鼠神经行为学及大脑的胆碱能系统的影响,并探讨其机制。方法以跳台成绩为依据,将ICR小鼠雌雄各63只随机分为空白组、模型组、安理申组及参知健脑胶囊超大、大、中、小剂量组,每组18只。各给药组给予相应药物灌胃,空白组、模型组给予等体积生理盐水。灌胃7 d后,皮下注射东莨菪碱4.5 mg/kg致获得性记忆障碍小鼠模型,之后予避暗实验,记录避暗被动躲避训练中各组动物的潜伏期、电击次数。取右侧大脑,比色法检测乙酰胆碱酯酶及乙酰胆碱转移酶活性。结果与模型组比较,大、中、小剂量组潜伏期差异有统计学意义(P<0.05),而超大剂量组、安理申组差异均无统计学意义;与模型组比较,除大剂量组外,各治疗组乙酰胆碱酯酶活性差异均有统计学意义(P<0.05),乙酰胆碱转移酶活性差异均无统计学意义。结论参知健脑胶囊可以抑制乙酰胆碱酯酶活性、增加乙酰胆碱转移酶活性,以中剂量的治疗作用较为稳定。Objective To observe the effects of Shenzhijiannao capsule on neuroethology and cerebral cholinergic system of acquired memory disorder model in mice.Methods According to the scores of step down test,ICR mice were randomly divided into seven groups,including control group,model group,aricept group,oversized large-,large-,medium-,small-dose Shenzhijiannao capsule group,with 18 mice in each group.The treatment group was given corresponding drug respectively,and control group and model group were given NS.After 7 d administration,acquired memory disorder model was made with scopolamine subcutaneous injection in the dose of 4.5 mg/kg.Then,the latency and the number of electric shock in passive avoidance test were recorded.After that,the activity of acetylcholinesterase（AchE） and acetylcholine transferase（AchT） of the right side of mice brain were measured with colorimetry.Results In the aspect of neuroethology,large-dose group,medium-dose group,small-dose group had siginificant difference compared with control group（P0.05）,but aricept group and oversized large-dose group had no difference.In the aspect of the activity of AchT,there was no difference between treatment groups and control group.In the aspect of the activity of AchE,except large-dose group,there was siginificant difference between treatment groups and control group（P0.05）.Conclusion Shenzhijiannao capsule may depress the activity of AchE and increase the activity of AchT,and the medium does of which have more stable effect.
