摘 要:目的研究磷酸肌酸钠对高位脊髓损伤后心肌细胞超微结构的影响。方法健康雄性SD大鼠30只,随机分为5组:假手术组(TC)、SCI损伤12h组(TS-T1)和24h组(TS-T2)、SCI损伤12h和24h后分别向腹腔一次性注射磷酸肌酸钠(TL-T1组和TL-T2组)。制备高位脊髓损伤模型。在各个时点抽血测定肌酸激酶(CK)和肌酸激酶同工酶(CK-MB)含量;透射电镜下观察大鼠右心尖心肌组织细胞超微结构。结果 TS组、TL组各时点血清CK和CK-MB含量均高于TC组(P<0.05);TL组CK和CK-MB含量低于TS组同一时点的含量(P<0.05)。透射电镜结果显示TC组各时点心肌超微结构正常,TS组各时点均出现心肌损伤的超微结构改变,TL组各时点心肌细胞损伤情况较TS组明显减轻。结论磷酸肌酸钠可减轻高位脊髓损伤后心肌细胞超微结构的损伤改变。Objective To investigate the effect of phosphocreatine on the ultrastructure of myocardial cell after high-level spinal cord injury(SCI).Methods 30 healthy male SD rats were randomly divided into five groups: Sham operation group(TC),SCI 12 h group(TS-T1) and SCI 24 h group(TS-T2),TL-T1 and TL-T2 groups with abdominal injection with phosphocreatine(200 mg/kg) after SCI 12 h and 24 h respectively.High-level spinal cord injury model was prepared.At each time point,blood creatine kinase(CK) and creatine kinase isoenzyme(CK-MB) levels were measured,and myocardial ultrastructure was studied using the transmission electron microscope(TEM).Results At each time point,blood CK and CK-MB levels in TS and TL groups were higher than those in TC group(P0.05),while in TL groups they were lower than those in TS groups(P0.05).ITEM results showed that at each time point,myocardial ultrastructures were normal in TC group while injury induced myocardial ultrastructure changes were observed in TS groups,but myocardial injury in TL group was significantly less severe than that in TS group.Conclusion In SCI rats,phosphocreatine can alleviate injury induced myocardial ultrastructure changes.
