摘 要:目的探讨氨溴索(AMB)协同万古霉素对金黄色葡萄球菌(SAU)生物被膜(BF)的破坏作用。方法构建SAU生物被膜体外模型;两倍稀释法测定最低抑菌浓度;扫描电镜(SEM)观察载体表面BF形态;MTT法行活菌计数。结果 3d或7d的BF,经AMB作用24h后用SEM观察,可见BF均明显少于对照组;BF内活菌计数显示,3d万古霉素(VAN)组、AMB+VAN组的SAU活菌计数分别为(19.14±5.64)、(6.44±1.53)、(3.13±0.95)×107 CFU/ml,7d分别为(43.28±3.74)、(27.39±4.97)、(17.61±2.14)×107 CFU/ml,均明显少于对照组(P<0.01),且AMB+VAN组更少于VAN组(P<0.01)。结论 AMB能破坏SAU已经形成的BF,与VAN联合应用后,可使BF内SAU显著减少,显示出了协同杀菌作用。OBJECTIVE To explore the destructive effect of ambroxol coordinated with vancomycin on the biofilm of Staphylococcus aureus.METHODS The in vitro model of S.aureus biofilm was constructed;two-fold dilution method was adopted to assay the minimum inhibitory concentration;the scanning electron microscope(SEM)was used to observe the biofilm;MTT assay was used to estimate the bacterial counts.RESULTS As for the 3 d or 7 d biofilm,after being reacted with ambroxol for 24 hours,the biofilm of the ambroxol group was significantly less than the control group observed by SEM,biofilm bacterial counts indicated that the bacterial counts of S.aureus under the effect of vancomycin for 3d and under the effect of ambroxol plus vancomycin were(19.14±5.64)×107CFU/ml,(6.44±1.53) ×107CFU/ml and(3.13±0.95)×107CFU/ml,(43.28±3.74)×107CFU/ml,(27.39±4.97),and(17.61±2.14)×10^7CFU/ml under the effect for 7 d,which were significanty less than the control group(P〈0.01),the bacterial counts of the ambroxol plus vancomycin group was significantly less than that of the vancomycin group(P〈0.01).CONCLUSION Ambroxol can destroy the biofilm having been formed by S.aureu,as combined with vancomycin,it can significantly reduce S.aureus inside the biofilm,which demonstrates the synergetic bactericidal effect.
