摘 要:目的探讨幼年大鼠单侧输尿管结扎所致肾间质纤维化模型中,波形蛋白和α-SMA的表达趋势及其与肾间质纤维化的相关性,同时观察螺内酯干预治疗后的效果。方法采用单侧输尿管结扎的方法制备肾间质纤维化大鼠模型(UUO模型),将96只SD雄性大鼠随机分为假手术组、模型组、螺内酯治疗组。造模成功后分别在7、14、21、28d每组各取8只大鼠处死,左肾组织行Masson染色,动态观察各组肾间质损伤程度,并用免疫组织化学链霉亲和素-生物素-过氧化物酶复合物(SABC)法检测各组Vimentin和α-SMA的表达水平。结果 随着时间的推移,模型组和治疗组肾间质损伤程度逐渐增强,但治疗组损伤较模型组轻微,二者的差异有统计学意义(P<0.01);在模型组中,波形蛋白和α-SMA的表达水平随肾间质损伤程度的加重而逐渐增强,波形蛋白各时间点之间差异有统计学意义(P<0.01),肾间质纤维化程度与波形蛋白和α-SMA的表达水平呈正相关(r=0.647,P<0.01);模型组中波形蛋白和α-SMA的表达水平高于治疗组,差异有统计学意义(P<0.01)。结论波形蛋白和α-SMA的异常表达可能参与了肾间质纤维化过程;螺内酯在改善肾间质纤维化的过程中起着一定作用,其机制中可能有波形蛋白和α-SMA的参与。Objective To explore the expression trends of Vimentin and α-SMA in rat models with unilateral ureteral obstruction (UUO)-induced renal interstitial fibrosis and their correlations with renal interstitial fibrosis, and the efficacy of Spirolactone intervention was observed as well. Methods UUO method was used to prepare the rat models with renal interstitial fibrosis. A total of 96 SD male rats were randomly divided into Sham-operated, UOO model and Spirolactone-treated groups. Thereafter, 8 rats were sacrificed in each group at 7, 14, 21 and 28 days after success modeling and performed Masson staining in tissues of left kidney in order to dynamically observe the degrees of interstitium injury. Moreover, the expression levels of Vimentin and α-SMA were detected in each group using immunohistochemical SABC method. Results As time went by, the degree of interstitium injury was gradually enhanced in model and treatment groups. Nevertheless, the degree of injury in treatment group was much milder than that in model group (P0.01). In model group, the expression levels of Vimentin and α-SMA were gradually increased along with the aggravated interstitium injury. Notably, significant difference was found in Vimentin in every time spots (P0.01). The degree of renal interstitial fibrosis was positively correlated with the expression levels of Vimentin and α-SMA (r=0.647, P0.01). The expression levels of vimentin and α-SMA in model group were higher than those in treatment group (P0.01). Conclusion The abnormal expressions of Vimentin and α-SMA may be due to their participations in the process of renal interstitial fibrosis. Spironolactone played a certain role in improving the process of renal interstitial fibrosis and its mechanism may be involved with the participations of Vimentin and α-SMA.
