富组蛋白1促进创面愈合的实验研究

摘　　要：[目的]探讨富组蛋白对表皮创面愈合的影响,为临床用药和创面护理提供新的思路和方向。[方法]通过细胞划痕试验,分为Hist1实验组、空白对照组、Hist1+丝裂霉素实验组、空白+丝裂霉素对照组,用丝裂霉素抑制细胞增殖,建立体外创面模型,于0h、16h、24h分别测量划痕区1.4 mm2内的愈合率。[结果]大约1.4mm2的创面16h愈合率为64.60%,24h愈合率为71.42%;而空白对照组16h愈合率为41.23%,24h愈合率为48.11%,两组相比,差异有统计学意义(P<0.05);而加入丝裂霉素后,16h愈合率为61.97%,明显高于空白+丝裂霉素对照组(32.25%)。[结论]30μg/mL生理浓度的富组蛋白1能明显促进表皮细胞增殖和迁移,且随着时间延长愈合率增高;提示富组蛋白1能促进非口腔上皮创面愈合。Objective：To probe into the influence of histatins 1 on epidermal wound healing,so as to provide the new thinking and directions for clinical medication and wound care. Methods：The cases were divided into Hist 1 group, blank control group, Histl 4- mitomyein experimental group,blank 4- mitomycin control group through cell scratch test, using mitomycin to inhibit cell proliferation and establish in vitro wound model and measure the healing rate in 1 mm scratch area respectively at the 0 h, 16 h,24 h. Results：The healing rate of 1.4 mm2 wound was 64.60% after 16 h,71.42% after 24 h;while the healing rate of blank control group was 41.23% after 16 h,was 48. 11% after 24 h,and the difference was statistically significant between two groups （P〈 0. 05） ; however, after adding mitomycin, healing rate was 6l. 97 % after 16h,it was significantly higher than that of blank group （32. 25%）. Conclusion：30 μg/mL physiological concentrations of histatins 1 can obviously promote epidermal cell proliferation and migration, and the healing rate increased with the prolongation of time,it prompted the histatinl can promote the nonoral epithelial wound healing.
