摘 要:目的:研究淫羊藿、补骨脂和骨碎补3种补肾壮骨中药对大鼠乳腺癌骨转移模型骨痛和骨质破坏的影响。方法:使用雌性SD大鼠进行实验,将48只大鼠随机分为6组,即假手术组、模型对照组、淫羊藿组、补骨脂组、骨碎补组和唑来膦酸组,其中后5组为手术组,在左侧胫骨髓腔内注射MRMT-1大鼠乳腺癌细胞建立大鼠乳腺癌骨转移模型,分别给予对应药物。在造模后第20天观察Von Frey纤维刺激的50%缩足阈(50%PWT)及双足负重差异以显示疼痛行为。第21天取材,测定肿瘤体积、左胫骨影像评分、骨矿物质含量(BMC)和骨密度(BMD)。结果:3种补肾壮骨中药均可使乳腺癌骨转移大鼠50%PWT提高(P<0.05或P<0.01),补骨脂可使双足负重差异减小(P<0.01),淫羊藿可使BMC和BMD升高(P<0.05和P<0.01)。此外,3种中药均有抑制肿瘤生长和减低骨破坏影像评分的趋势,但未见统计学差异。结论:淫羊藿等补肾壮骨中药在大鼠乳腺癌骨转移模型上具有减轻癌性骨痛和骨质破坏的作用。Objective: To study effects of TCM herbs with function of reinforcing kidney and strengthening bones on tumor pain and bone destruction in a rat model of bone metastasis from breast cancer.Methods: Sprague-Dawley rats receiving intra-tibial injections of syngeneic MRMT-1 rat mammary gland carcinoma cells developed bone metastasis.To observe the effects,Pure water,Epimedium brevicornum,Fructus psoraleae,Rhizoma drynariae and Zoledronic acid were given to different groups respectively.On the 20th day after MRMT-1 cell injection,50% Paw withdrawal threshold(50%PWT) and the difference of weight bearing between hind paws were tested in all groups.On the 21st day,all animals were killed and volume of tumor were observed.The development of bone structural damage to the bone was measured by radiological analysis,quantitative measurement of bone mineral density(BMD) and bone mineral capacity(BMC) was monitored by dual energy X-ray bone density measurement.Results: The three kinds of herbs could increase 50%PWT(P〈0.05 or P〈0.01),the difference of weight bearing between hind paws was reduced by Rhizoma drynariae,and BMD and BMC were enhanced by Epimedium brevicornum(P〈0.05 or P〈0.01).Conclusion: Traditional Chinese herbs of reinforcing kidney and strengthening bones can alleviate pain from bone tumor and inhibit bone destruction induced by tumor.
