摘  要:[目的]为野生小花碧冬茄组织快繁技术提供依据,文中研究了组培条件下,不同浓度的6-BA、NAAI、BA处理对野生小花碧冬茄外植体器官分化和植株再生的影响。[方法]以野生小花碧冬茄无菌种子获得的试管苗为材料,筛选其成苗培养和生根培养最适激素组合。[结果]成苗培养中,较高浓度的6-BA配合较低浓度的NAA有利于外植体形成丛生芽,而高水平的NAA会引起愈伤组织的大量产生。对野生小花碧冬茄而言,芽分化最佳培养基组合为MS+蔗糖30 g/L+琼脂粉5 g/L+NAA 0.2 mg/L+6-BA 1.5 mg/L;愈伤组织最佳培养基组合为MS+蔗糖30 g/L+琼脂粉5 g/L+NAA 0.4 mg/L+6-BA 1.0 mg/L;生根最佳培养基组合为1/2MS+蔗糖30 g/L+琼脂粉5 g/L+IBA 1.0 mg/L+NAA 0.4 mg/L。[结论]明确野生小花碧冬茄芽分化、愈伤组织和生根培养的理想激素配比,初步建立了野生小花碧冬茄的高效再生体系。[ Objective ] This study aimed to provide the basis for rapid propagation technology of wild Petunia Juss.. [ Method ] Effects of dif- ferent plant hormones 6-BA, NAA and IBA on the organ differentiation and plantlet regeneration of wild Petunia Juss.. explants were investi- gated. The optimal medium for seedling culture and rooting induction were selected with aseptic plantlets of wild Petunia Juss.. [ Result ] Higher concentration of 6-BA in combination with lower concentration of NAA is beneficial to the formation of multiple-bud clumps induced by callus, and a higher level of NAA will lead to a sharp increase of callus. The optimal medium for bud differentiation is MS + 30 g/L of sucrose + 5 g/L of agar +0.2 mg/L of NAA + 1.5 mg/L of 6-BA; the optimal medium for callus culture is MS +30 g/L of sucrose +5 g/L of agar + 0.4 mg/L of NAA + 1.0 mg/L of 6-BA ; the optimal medium for rooting induction is 1/2MS + 30 g/L of sucrose + 5 g/L of agar + 1.0 mg/L of IBA + 0.4 mg/L of NAA. [ Conclusion] The optimal exogenous hormone combinations for bud regeneration, callus culture and rooting induc- tion were defined, an efficient regeneration system of wild Petunia Juss.. was established preliminarily.
