大鼠供肝质量的快速评价方法

摘　　要：目的　探讨供肝质量的快速评价方法。方法　建立大鼠酒精性肝病模型 ,然后取不同时期的大鼠肝组织做快速石蜡切片、普通石蜡切片和冰冻切片 ,观察病理改变 ,并行病理学综合评分 ,同时对肝脏进行门静脉灌洗 ,监测灌洗时门静脉压力及流量。结果　实验组病理学综合评分随实验时间的延长逐渐增高 ;快速石蜡切片的病理学综合评分与普通石蜡切片比较 ,差异无显著性 (P >0 .0 5 ) ;实验组大鼠在恒定流量进行门静脉灌洗时 ,门静脉压力升高 ,与实验对照组比较 ,差异有显著性 (P <0 .0 5 ) ,且门静脉压力变化与病理学综合评分呈高度正相关 (r =0 .819,P <0 .0 0 1)。结论　快速石蜡切片与普通石蜡切片一样 ,可对供肝病理学改变做出准确评估 ;在门静脉灌洗液流量恒定的情况下 ,门静脉压力与供肝的病理学改变高度相关 ;快速石蜡切片和灌洗时门静脉压力测定相结合 。Objective To explore a rapid method for evaluating the quality of donor liver by biopsy and flush of portal vein perfusion. Methods Rat alcoholic liver disease model was established. Rapid para ffin sections, common paraffin sections and frozen sections were made at different pathological stages of the liver. The pathological changes were observed. Liver portal perfusion was performed to monitor the portal pressure and flow. Results In the experimental group, pathology score was gradually increased with the delay of experimental time. There was no significant difference in pathology score between rapid paraffin sections and common paraffin sections ( P > 0.05 ). There was significant difference in the portal pressure between the experimental group with stable flow for portal perfusion and experimental control group ( P < 0.05 ). There was a positive relation between portal vein pressure and pathology score ( r = 0.819 , P < 0.001 ). Conclusions Both rapid paraffin sections and common paraffin sections can accurately evaluate the pathological changes of the donor liver. There existed a positive relation between portal vein pressure and liver biopsy. Combination of rapid paraffin sections and portal vein pressure can evaluate the quality of donor liver correctly.
