摘  要:[目的]建立一种测定植物中拌种灵残留的HPLC分析方法。[方法]以烟草为供试植物,利用甲醇浸提、固相萃取法净化进行了样品前处理,并在醇∶水=55∶45(V/V)为流动相、流速1.0 ml/min、进样量20μl、紫外检测波长270 nm的条件下采用HPLC法(ODS-C18柱)测定了样品中拌种灵的残留量。[结果]在该条件下,拌种灵的保留时间为7.20 min,峰面积关于标准溶液浓度(0.1~100.0μg/ml)的回归方程为y=71.31x+76.26,相关系数为0.999 9,回收率在90.30%~97.93%,变异系数在1.26%~6.63%。[结论]该方法的回收率、精密度均能完全满足残留分析要求,具有一定的适用性。[ Objective ] The aim was to establish a residue analysis method of seedvax in plants by HPLC. [ Method ] The tabacco seedlings were extracted with methanol, cleaned with SPE-C18 and separated by ODS-C18 column, and then the residual of seedvax in tabacco seedlings was determined by HPLC under conditions of the mobile phase 55% (V/V) methanol with flow velocity 1.0 ml/min, the sample volume 20μl and the UV detector wavelenghth 270 rim. [ Result] Under the conditions, the reserved time of seedvax was 7.20 rain ; the peak area showed good linearity with the corresponding concentration standard solution of seedvax within the range of 0.1 - 100.0 μg/ml ( r = 0. 999 9 ). The re- gression calibration curve equation was y =71.31x +76.26. The recovery was from 90.30% to 97.93%, and the coefficient of variation was from 1.26% to 6.63%. [ Conclusion] The HPLC analysis method of seedvax residue has good precision and accuracy, so it has certain suitability.
