摘 要:斑潜蝇隶属双翅目(Diptera)潜蝇科(Agromyzidae)斑潜蝇属(Liriomyza),是一类重要的农业害虫。近年来,随着海峡两岸果蔬贸易的快速发展,斑潜蝇类害虫传入大陆地区的风险明显增加,植物检疫部门急需该类害虫的快速鉴定技术和方法。本研究针对我国进境检疫性害虫三叶草斑潜蝇(L.trifolii),在检测和检索其线粒体DNA细胞色素氧化酶I(mtDNA COI)基因序列并与其近缘种美洲斑潜蝇(L.sati-vae)和南美斑潜蝇(L.huidobrensis)相同基因序列比对的基础上,设计并筛选了其种类识别特异引物,初步实现了三叶草斑潜蝇的快速鉴定。Abstract Liriomyza is a kind of agricultural insect pest which belongs to Diptera, Agromyzidae. With rapid de- velopment of vegetable and flower trades between Taiwan and mainland, the entry risk of Liriomyza increases obvi- ously. The rapid methods and techniques of quarantine identification for Liriomyza are needed urgently at ports. A specific primer pair of L. trifolii( Burgess), the quarantine pest of Liriomyza, were studied in this study. Basing on testing, collecting and comparing the sequences of mitochondrial cytochrome c oxidase subunit I (mtDNA COI) of L. trifolii ( Burgess), L. sativae Blanchard and L. huidobrensis ( Blanchard), the proper specific primer pair was designed and selected which could accomplish the rapid identification of L. trifolii(Burgess) initially.
