摘 要:为分析我国猪瘟病毒(CSFV)流行株的抗原性差异及其与特异性单克隆抗体(MAb)反应性之间的关系,本研究采用CSFV强弱毒鉴别MAb通过ELISA方法对我国流行的29株分离株进行抗原反应性研究,结果显示针对弱毒株MAb(MAb11)除3株为阳性外,其余均为阴性;而这些分离株可以被强毒MAb(MAb56)分为强反应和弱反应两组。同时对29株分离株E2基因的190 nt进行同源性分析,并将其分为3个亚群,即1.1、2.1和2.2;氨基酸序列具有一致独特的位点差异规律,强反应组(761G、764L)和弱反应组(761R或K、764P)分别具有2个独特的差异位点。推测761G和764L是针对MAb56与CSFV抗原结合的关键位点。To investigate antigenic relationship of classical swine fever virus (CSFV) isolates, a total of 29 CSFV field isolates collected from different areas in China were tested by ELISA established with differential monoclonal antibodies (MAb) of MAbl 1 specific to the attenuated CSFV vaccine strain (HCLV) and MAb56 specific to the virulent CSFV. Three field isolates were recognized by MAbll. The field isolates were able to be classified into 2 groups according to the reactivity with MAb56, designated strong reactivity (SR) group and weak reactivity (WR) group. The field isolates were clustered within group 2 and subdivided into three subgroups (1.1, 2.1, 2.2), which revealed that there were 2 unique mutations site found in 2 groups (761^G and 764^L in SR group, while 761^R and 764^p in WR group). It was suggested that 761^G and 764L were the residues determining the binding of the viruses with MAb56.
