摘 要:目的建立甘草饮片HPCE-DAD指纹图谱分析方法,并对甘草及其炮制品的指纹图谱进行比较。方法采用高效毛细管电泳法进行色谱分离,以40 mmol.L 1硼砂-10 mmol.L 1磷酸二氢钠-10%甲醇(pH=8.6)为运行缓冲液,分离电压为20 kV,波长为254 nm,以甘草酸为参照物(IS),测定其指纹图谱,并作模糊聚类法分析和相似度评价。结果初步建立了以10个共有峰为特征指纹信息的甘草饮片HPCE-DAD指纹图谱,发现少数甘草饮片的指纹图谱有一定差异,生品与其炮制品的指纹谱中共有峰相对峰面积差异显著。结论本方法准确可靠,重现性好,可作为甘草饮片内在质量评价的依据。OBJECTIVE To establish the analytical method for the fingerprint of Glycyrrhizae Radix et Rhizoma by HPCE-DAD and compare the fingerprints of Glycyrrhizae Radix et Rhizoma and its processed products.METHODS Based on the mode of high performance capillary electrophoresis,40 mmol·L 1 sodium borate-10 mmol·L 1 sodium dihydrogen phosphate-10% methanol(pH 8.6) was used as buffer solution.The separation voltage was 20 kV and the detection wavelength was set at 254 nm.Glycyrrhizic acid was used as reference standard,the chromatographic fingerprint were determined.The data were analyzed by fuzzy cluster and fingerprint similarity evaluation software to compare the similarity of samples.RESULTS HPCE-DAD fingerprints with 10 common peaks of Glycyrrhizae Radix et Rhizoma were established preliminarily.It was discovered that a small number of samples differed from others.Regarding to the fingerprints of Glycyrrhizae Radix et Rhizoma and its processed products,there were obvious differences in the relative areas of common peaks.CONCLUSION The method is reliable,accurate and can be used for quality control of Glycyrrhizae Radix et Rhizoma.
